Transnovasomes is a new and exciting class of lipid-based nanovesicles. Colchicine (COL) hydrophilic natural alkaloid with anti-inflammatory anti-mitotic features having oral administration permeation defects. Recurrent Aphthous Ulcer (RAU) the most prevalent disease mucosa suffering from lack particular final preventative therapy. So, designing prolonged effective specialized delivery system for ulcer treatment important. transnovasomes (COL-TNs) were prepared using surfactants (Span 60 & Span 80), free fatty acids (Oleic acid Stearic acid), Cholesterol Brij 58. The COL-TNs evaluated their vesicle size (VS), polydispersity index (PDI), zeta potential (ZP), entrapment efficiency (EE%), ex-vivo after 12 hours (Q12h). Values VS, PDI, ZP, EE% Q12h optimized formulation 256.74 ± 11.2 nm, 0.322 0.08, -43.3 0.62, 85.35 3.7% 72.69 5.84% respectively. Drug accumulation was ninefold greater than drug solution 8 hours. In-vivo, formulation, alone or in combination platelet-rich plasma (PRP), achieved complete healing acetic-acid induced RAU restoring normal levels assayed biomarkers (TNF- α, NF-κβ, IL-6, MMP9, VEGF) histological mucosa. In conclusion, can be used as promising, safe, efficient RAU, monotherapy therapy PRP.
