Abstract Streptococcus mutans is a commensal bacterium in the oral cavity, but its overgrowth leads to biofilm formation and dental caries. S. glucan-binding protein C (GbpC) cornerstone of scaffold thus rational target for inhibition. Here we present selection DNA aptamers GbpC aid development inhibiting drugs. During SELEX, used extracellular domain as structurally homologous antigen I/II GbpC-deficient strain counter-targets. The aptamer candidates obtained were panned using method based on primer-blocked asymmetric PCR AlphaScreen. interaction between most promising panning was analysed by biolayer interferometry microscale thermophoresis. Finally, tested inhibitory effect wild-type . measurements demonstrated both efficacy selectivity aptamers. Two studied reduced 30% case strains no reduction detected strain, confirming suitability strategy presented providing lead molecules inhibitors.
