Lipid nanoparticles (LNPs) are widely recognized as effective drug delivery systems for RNA therapeutics because their efficacy is critically dependent on structural organization. The mesoscopic architecture of these multicomponent systems, which governed by interactions among ionizable lipids, nucleic acids, and stabilizers, dictates encapsulation efficiency, biodistribution, therapeutic performance. Although small-angle X-Ray scattering (SAXS) enables nanostructure characterization, the absence suitable analytical models has hindered LNP development. Here, we present a core-triple shell SAXS model that resolves hierarchical organization, including inner lipid layer, intermediate hydrophilic region, outer PEG corona. For LNPs encapsulating mRNA, Gaussian distribution was implemented to characterize quasi-periodic structure originating from self-assembly mRNA-ionizable complexes. Validation studies employing Comirnaty-based demonstrated controlled variation nitrogen-to-phosphorus (N/P) ratios produced distinguishable features establish quantitative correlations between N/P assembled structure. modeling framework provides pharmaceutical researchers with robust tools systematic stability assessment precision formulation optimization LNPs. These insights expected advance development next-generation potentially enhancing efficiency pharmacokinetic properties.
