Abstract
The
spatial
and
temporal
changes
of
the
COVID-19
pandemic
have
been
monitored
with
wastewater-based
surveillance,
which
many
countries
applied
to
their
national
public
health
monitoring
measures.
most
commonly
used
methods
for
detection
SARS-CoV-2
in
wastewater
are
RT-qPCR
RT-ddPCR.
Previous
comparisons

A
focus
on
water
quality
has
intensified
globally,
considering
its
critical
role

sustaining
life

ecosystems.
Wastewater,
reflecting
societal
development,
profoundly
impacts
public
health.
Wastewater-based
epidemiology
(WBE)
emerged
as
a
surveillance
tool
for
detecting
outbreaks
early,
monitoring
infectious
disease
trends,
providing
real-time
insights,
particularly
vulnerable
communities.
WBE
aids
tracking
pathogens,
including
viruses,
sewage,
offering
comprehensive
understanding
of
community
health
lifestyle
habits.
With
the
rise
global
COVID-19
cases,
gained
prominence,
aiding
SARS-CoV-2
levels
worldwide.
Despite
advancements
treatment,
poorly
treated
wastewater
discharge
remains
threat,
amplifying
spread
water-,
sanitation-,
hygiene
(WaSH)-related
diseases.
WBE,
serving
complementary
surveillance,

pivotal
community-level

Molecular
analysis
of
public
wastewater
has
great
potential
as
a
harbinger
for
community
health
and
threats.
Long-used

Objective:
The
aim
of
this
study
is
to
evaluate
the
effectiveness
three
commercial
nucleic
acid
extraction
kits
(kit
A,
B
and
C)
in
isolating
SARS-CoV-2
viral
RNA
from
wastewater
samples.
Method:
In
study,
water
samples
were
collected
March
2021
treatment
plants
located
different
parts
Istanbul,
it
was
confirmed
that
they
negative
for
SARS-CoV-2.
Different
concentrations
virus,
previously
inactivated
at
BSL-3
laboratory
Pendik
Veterinary
Control
Institute,
added
quantification
performed
using
RT-qPCR
kit
specific
Results:
At
end

yielded
highest

consistent
results,
significantly
outperforming
other
two
