Abstract The spatial and temporal changes of the COVID-19 pandemic have been monitored with wastewater-based surveillance, which many countries applied to their national public health monitoring measures. most commonly used methods for detection SARS-CoV-2 in wastewater are RT-qPCR RT-ddPCR. A focus on water quality has intensified globally, considering its critical role in sustaining life and ecosystems. Wastewater, reflecting societal development, profoundly impacts public health. Wastewater-based epidemiology (WBE) emerged as a surveillance tool for detecting outbreaks early, monitoring infectious disease trends, providing real-time insights, particularly vulnerable communities. WBE aids tracking pathogens, including viruses, sewage, offering comprehensive understanding of community health lifestyle habits. With the rise global COVID-19 cases, gained prominence, aiding SARS-CoV-2 levels worldwide. Despite advancements treatment, poorly treated wastewater discharge remains threat, amplifying spread water-, sanitation-, hygiene (WaSH)-related diseases. WBE, serving complementary surveillance, is pivotal community-level viral infections. However, there untapped potential to expand surveillance. This review emphasizes importance link between health, highlighting need further integration into management.

Molecular analysis of public wastewater has great potential as a harbinger for community health and threats. Long-used to monitor the presence enteric viruses, in particular polio, recent successes reliable lead indicator trends SARS-CoV-2 levels hospital admissions generated optimism emerging evidence that similar science can be applied other pathogens pandemic (PPPs), especially respiratory viruses their variants concern (VOC). However, there are substantial challenges associated with implementation this ideal, namely multiple distinct fields inquiry must bridged coordinated. These include engineering, molecular sciences, temporal-geospatial analytics, epidemiology medical, governmental messaging, all which present own caveats. Here, we outline framework an integrated, state-wide, end-to-end human pathogen monitoring program using track viral PPPs.

The post-pandemic world still faces ongoing COVID-19 infections, although international travel has returned to pre-pandemic conditions. Wastewater-based epidemiology (WBE) is considered an efficient tool for the population-wide surveillance of infections during pandemic. However, performance WBE in era with restrictions lifted remains unknown. Utilizing weekly county-level wastewater data from June 2021-November 2022 222 counties 49 states (covering 104 million people) United States America, we retrospectively evaluated correlations between SARS-CoV-2 RNA (C

Objective: The aim of this study is to evaluate the effectiveness three commercial nucleic acid extraction kits (kit A, B and C) in isolating SARS-CoV-2 viral RNA from wastewater samples. Method: In study, water samples were collected March 2021 treatment plants located different parts Istanbul, it was confirmed that they negative for SARS-CoV-2. Different concentrations virus, previously inactivated at BSL-3 laboratory Pendik Veterinary Control Institute, added quantification performed using RT-qPCR kit specific Results: At end determined C yielded highest total produced more consistent results, significantly outperforming other two terms yield purity. Statistical analysis revealed significant differences (p < 0.05) gene copy numbers 0.01) between kits, demonstrated superior linearity reproducibility. Conclusion: According findings, although all evaluated are suitable detecting samples, provides most efficient reliable performance, especially high-throughput studies. Additionally, highlights importance selecting appropriate methods surveillance, which serves as an early warning system outbreaks threaten public health.
