This study investigated the protective effects and mechanisms of cow placenta peptides (CPP) on intestinal barrier damage in aging model mice. Forty-eight male ICR mice were assigned to four groups: a control group (N), an (M), CPP treatment (T), vitamin C (P). Groups T P received oral administration (2000 mg/kg/day) (100 mg/kg/day), respectively, while groups M, T, subjected intraperitoneal injections D-galactose (D-gal) (300 mg/kg/day). Group N equivalent volume normal saline via injection. Treatments administered once daily for 8 weeks. The results demonstrated that significantly alleviated D-galactose-induced structural damage, increasing villus height-to-crypt depth ratio reducing serum diamine oxidase (DAO) lipopolysaccharide (LPS) levels. notably oxidative stress inflammation, restored tight junction expression, enhanced integrity. Transcriptome sequencing identified 1396 DEGs associated with CPP’s effects, highlighting TLR4, IL-1β, Mmp9 as core regulatory genes through protein–protein interaction network analysis. Kyoto Encyclopedia Genes Genomes Gene Ontology enrichment analyses implicated TLR4/NF-κB signaling pathway, which was further validated. Western blotting confirmed down-regulated IKKβ, p-NF-κB p65 protein expression intestines In conclusion, effectively alleviates D-gal-induced by enhancing antioxidant defense inhibiting thereby diminishing inflammation protecting

Introduction Sheep placenta extract (SPE) is a representative traditional medicinal substance that exhibits multiple experimentally validated physiological properties, including anti-aging effects, wound healing acceleration, antioxidant activity, and anti-inflammatory mechanisms. However, the mechanism by which SPE influences delay of aging still not yet clear. Methods Exploring effects sheep on D-gal induced senescence in mouse model macrogenomics metabolomics. Results In serum mice treated with SPE, levels function such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) were notably higher compared to those blank group, whereas malondialdehyde (MDA) decreased. We revealed alleviated changes gut microbiota caused mice, significant decrease Firmicutes/Bacteroidetes (F/B) ratio gut. Furthermore, Akkermansia muciniphila ( A. ), known for its regulating immune response potential showed increase 1177.94%. The analysis UHPLC-QE-MS combined orthogonal partial least squares discriminant (OPLS-DA) screening differential metabolites metabolic profiles upregulation cis -5,8,11,14,17-eicosapentaenoic acid (EPA) triptolide metabolites, following treatment, are commonly believed have immunosuppressive, anti-inflammatory, anti-proliferative, anti-tumor effects. Discussion role ameliorating may be associated increased abundance accumulation two EPA triptolide, serum.

Our preliminary study identified dairy cow placenta extract (CPE) as a mixture of peptides with potent antioxidant activity both in vivo and vitro. However, the specific (AOPs) responsible for this were not yet identified. In current study, we employed virtual screening chromatography techniques to isolate two peptides, ANNGKQWAEVF (CP1) QPGLPGPAG (CP2), from CPE. These found be less stable under extreme conditions such high temperature, strong acid, alkali, simulated digestive conditions. Nevertheless, normal physiological conditions, CP1 CP2 exhibited significant properties, including free-radical scavenging, metal chelating, inhibition lipid peroxidation. They also up-regulated activities intracellular enzymes response hydrogen-peroxide-induced oxidative stress, resulting reduced MDA levels, decreased expression Keap1 gene protein, increased levels Nrf2 HO-1 genes proteins. Furthermore, demonstrated superior compared CP2. findings suggest that hold potential mitigating stress vitro highlight efficacy method isolating AOPs within

The global aging population has led to a rise in age-related health issues, such as malnutrition, metabolic disorders, and even immune decline. Among these concerns, periodontitis holds particular significance for the well-being of elderly. This study aimed investigate impact on inflammatory resorption alveolar bone mice with periodontitis, specific focus alterations intestinal microenvironment. To achieve this, we established D-galactose (D-gal)-induced mouse model employed histopathological staining, oxidative stress, factors analyses assess severity status. Additionally, 16S rRNA sequencing untargeted metabolomics analysis were microbiota metabolites. Our results showed that D-gal-induced experienced more pronounced disruptions gut barrier, accompanied by an overall decline physical condition. microbial composition structure aged also underwent significant modifications, decreased Firmicutes/Bacteroidetes (F/B) ratio. Furthermore, demonstrated primarily influenced lipids lipid-like molecules metabolism, enrichment observed rheumatoid arthritis histidine metabolism pathways. These findings provide further evidence process exacerbates loss (ABL) through disturbances homeostasis.

The risk of acquiring new intramammary infections is high at the end lactation, especially for milk-producing dairy animals. Resistance to bacterial infection increases following completion mammary gland involution after milking cessation. serotonin precursor 5-hydroxytryptophan (5-HTP) could accelerate by increasing circulating levels, but ruminal microbes may degrade 5-HTP if orally administered adult ruminants. It unclear whether rumen-protected effectively mediate (5-hydroxytryptamine, 5-HT) and therefore in Goats were used as a model current study investigate effects on behaviour, 5-HT metabolism, In first experiment, 16 female Dazu black goats assigned one four groups randomised block design. treatments included basal diet plus 0, 4, 20, or 100 mg/kg BW 5-HTP. Serum was collected 3, 6, 12, 24 h offering morning feed, behaviours monitored. second 12 (Somatic cell count < 250 000) randomly control (basal diet) group 20 BW). Milk secretions manually aseptically d -1, 1, 2, 5 around weaning. results depicted that supplementation elevated 5-hydroxyindole acetic acid concentrations, while lowered goats' locomotive activity. A concentration (100 BW) increased serum alkaline phosphatase gamma-glutamyl transpeptidase concentrations. Moreover, oral with accelerated reduced feed intake These demonstrate influences metabolism accelerates cessation
