Colitis, a troublesome inflammatory disease that significantly impacts daily life, has garnered considerable attention in recent times. Protolysates play crucial role the treatment of colitis, and intestines ovum Cucumaria frondosa represent readily available source these hydrolysates. However, effects C. hydrolysates (CFHs) on colitis have not been thoroughly investigated. We initially examined molecular weight distribution CFHs found fraction molecules with less than 1000 Da accounted for 86.98%, indicating hydrolysis primarily produced oligopeptides. Subsequently, we employed dextran sulfate sodium-induced experimental model to assess therapeutic potential CFHs. The findings indicated preventive administration dramatically attenuated pathological manifestations associated mice, including loss, colon shortening, tissue damage. Furthermore, suppressed secretion pro-inflammatory cytokines IL-6, TNF-α, IL-1β, as well MPO tissue. Introduction Cordyceps militaris extract (CME) and cordycepin (CPN) are biomolecules with a wide range of biological activities, including anti-inflammatory, antioxidant anti-tumour effects. The research objective wasto investigate the influences CME CPN on colonic morphology, microbiota composition metabolomics in lipopolysaccharide (LPS)-challenged piglets. Methods Twenty-four weaned castrated piglets were randomly divided into four groups: control group (fed basal diets), LPS CPN-LPS (basal diets + 60 mg/kg cordycepin), CME-LPS C. militari sextract). On 21st day, LPS, CPN-LPS, groups received an injection 100 μg/kg BW while was given sterile saline. Polysaccharides, vital biological macromolecules ubiquitous in organisms, have garnered attention as potential therapeutic candidates for ulcerative colitis (UC). However, the of Rhodiola crenulata polysaccharides (RCP) UC remains largely unexplored. The RCP was prepared by boiling water extraction, 80% alcohol precipitation, membrane separation, and D101 macroporous resin purification. monosaccharide composition (Mw = 67.848 kDa) includes mannose, rhamnose, glucuronic acid, galacturonic glucose, galactose, arabinose, with a molar ratio 0.22:1:0.07:7.03:2.88:0.64:4.12. In vivo experiments shown that can improve DSS induced weight loss mice, decrease disease activity index (DAI), alleviate histopathological changes colon tissue, suppress levels pro-inflammatory cytokine IL-6 MPO activity. Immunohistochemical results showed essential tight junction proteins such occludin, claudin1, ZO-1 were upregulated, improving integrity intestinal barrier. Importantly, regulated abundance microbiota reducing Firmicutes-to-Bacteroidetes (F/B), increasing beneficial bacteria Muribaculaceae Bifidobacterium , decreasing harmful including Erysipelotrichaceae, Faecalibaculum Lachnospiraceae_unclassified Parabacteroides Ruminiclostridium_9 . Additionally, it enhanced restoration acetic propionic isovaleric valeric acid to maintain SCFA levels, thereby restoring microecology.
