Introduction Yunvjian (YNJ) decoction, a classic traditional Chinese medicine prescription for inflammatory diseases, has demonstrated good therapeutic effects in the clinical treatment of pneumonia. The aim this study was to clarify effective ingredients and mechanism action YNJ on lipopolysaccharide (LPS)-induced acute lung injury (ALI). Methods were evaluated mouse model LPS-induced ALI LPS-treated MLE-12 murine epithelial cells RAW264.7 macrophages vitro . these systems studied using RNA sequencing, immunohistochemical analysis, immunoblotting, immunofluorescence, ELISA, polymerase chain reaction assays. Ultra-high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry applied identify absorbed components YNJ. Results attenuated pulmonary damage mice, as evidenced by reduced protein content bronchoalveolar lavage fluid, decreased wet/dry weight ratio, improved respiratory function. Analysis pneumonia-related samples from patients Gene Expression Omnibus dataset GSE40012 indicated that NOD-like receptor 3 (NLRP3)-mediated pyroptosis primary ALI. phosphorylation nuclear factor-kappa B (NF-κB) expression levels NLRP3, apoptosis-associated speck-like containing CARD (ASC), cleaved caspase-1, interleukin-1β (IL-1β) vivo Administration YNJ-containing serum increased cell viability malondialdehyde reactive oxidative species contents LPS-stimulated cells. also secretion tumor necrosis factor-α, IL-6, IL-1β macrophages, promoted macrophage polarization toward an M2 phenotype.
