As environmental plastic waste degrades, it creates an abundance of diverse microplastic particles. Consequently, microplastics contaminate drinking water and many staple food products, meaning the oral ingestion is important exposure route for human population. Microplastics have long been considered inert, however their ability to promote microbial dysbiosis as well gut inflammation dysfunction suggests they are more noxious than first thought. More alarmingly, there evidence permeating from throughout body, with adverse effects on immune nervous systems. Coupled now-accepted role gut-brain axis in neurodegeneration, these findings support hypothesis that this ubiquitous pollutant contributing rising incidence neurodegenerative diseases, like Alzheimer’s disease Parkinson’s disease. Microplastic (MP) are an emerging environmental pollutant, which has toxic effects on organisms, and it received extensive attention currently. Studying the transcriptomic metabolic responses of mice to nanoplastic-contaminated water is critical for understanding molecular-level toxicity nanoplastics (NPs), but there few studies this topic. To analyze different concentrations polystyrene (PS) at transcriptome metabolism spleens study molecular toxicity. Here, testing histopathology spleen female was performed after drinking containing 0.1 μm PS-NPs (1 mg/mL 50 mg/mL) 49 days, respectively. The tissue samples were subjected metabolome sequencing. Four differentially expressed genes randomly chosen qRT-PCR confirm correctness Common Kyoto encyclopedia genomes (KEGG) pathway analysis showed that a large number differential metabolites mainly focused immune, inflammation, neurodegenerative disease, cardiovascular nervous, etc. in organism systems module; lipid, amino acid, taurine hypotaurine metabolisms, signaling translation, molecules interaction, neuroactive ligand-receptor information processing. results levels confirmed immune system affected contaminated with nanoplastics.

Exposure of organisms to nanoplastics (NPs) is inevitable given their global abundance and environmental persistence. Nanoplastics (NPs) have been widely detected in freshwater environments and photodegradation, as well physical chemical breakdown, lead to different surface charges on the plastics. Although evidence literature highlights importance of NPs charge neurotoxicity, substantial gaps mechanistic understanding remain. In current study, zebrafish (Danio rerio) were exposed differentially charged (PS, PS-NH2, PS-COOH) at environmentally relevant concentration (10 μg/L). After full life cycle exposure, potential brain damage, altered metabolism was investigated through light sheet microscopy 3-dimensional imaging, histopathology, Evans blue dye (EBD) extravasation, gene expression, untargeted targeted metabolomics tissue zebrafish. Exposure PS, PS-COOH caused adverse effects performance neurobehaviors, blood-brain-barrier (BBB) permeability, amino acid metabolism, damage BBB mitochondria, overt inflammatory response. PS-NH2 (4.56-fold) (3.59-fold) accumulated reticular formation (RF) male brain, while only RF (6.57-fold) ventral hypothalamus (Hv) (3.08-fold) female brains. Several important biological pathways negatively impacted a charge- gender-specific fashion.
