Abstract
The
Japanese
encephalitis
virus
(JEV)
is
a
mosquito-borne,
enveloped
flavivirus
that
causes
acute
encephalitis.
Although
JEV
increasingly
recognized
as
global
threat,
there
currently
no
FDA-approved
treatment
available
for
JEV.
25-hydroxycholesterol
(25-HC)
an
oxysterol
produced
through
the
oxidation
of
cholesterol,
process
catalyzed
by
cholesterol
25-hydroxylase
(CH25H),
which
interferon-stimulated
gene
upregulated
during
viral
infections.
In
this
study,
we
report
first
time
25-HC
effectively
prevents
infection
in
cells.
Our
results
show
inhibits
Vero
cells
with
IC50
∼
3.7
μM,
while
displaying
cell
toxicity
(CC50
>
50
μM).
Given
antiviral
effects
25HC
against
Zika
and
Dengue
viruses,
our
findings
suggest
could
be
promising
candidate
development
broad-spectrum
anti-flaviviral
therapeutics.

Lipid
droplets
(LDs),
serving
as
the
convergence
point
of
energy
metabolism
and
multiple
signaling
pathways,

garnered
increasing
attention

recent
years.
Different
cell
types
within
central
nervous
system
(CNS)
can
regulate
to
generate
or
degrade
LDs
response
diverse
pathological
stimuli.
This
article
provides
a
comprehensive
review
on
composition
CNS,
their
generation
degradation
processes,
interaction
mechanisms
with
mitochondria,
distribution
among
different
types,
roles
played
by
these
cells-particularly
microglia
astrocytes-in
various
prevalent
neurological
disorders.
Additionally,
we
also
emphasize
paradoxical
role
post-cerebral
ischemia
inflammation
explore
potential
underlying
mechanisms,
aiming
identify
novel
therapeutic
targets
for
this
disease.

can
mediate
liver

either
directly
by
influencing
cholesterol
levels
or
through
non-cholesterol
pathways.
These
pathways,
which
operate
independently
cholesterol’s
traditional
metabolic
functions,
are
regulated

transcription
factors,
receptors.
Dysregulation
