The occurrence of obesity and related metabolic disorders is rising, necessitating effective long-term weight management strategies. With growing interest in the potential role gut microbes due to their association with responses different loss diets, understanding mechanisms underlying interactions between diet, microbiota, remains a challenge. This study aimed investigate impact multiphase dietary protocol, incorporating an improved ketogenic diet (MDP-i-KD), on microbiota. Using metagenomic sequencing, we comprehensively analyzed taxonomic functional composition microbiota 13 participants before after 12-week MDP-i-KD intervention. results revealed significant reduction BMI (9.2% loss) among obese following Machine learning analysis identified seven key microbial species highly correlated MDP-i-KD, Parabacteroides distasonis exhibiting highest response. Additionally, co-occurrence network post-weight-loss demonstrated healthier state. Notably, pathways nucleotide biosynthesis, aromatic amino acid synthesis, starch degradation were enriched pre-intervention positively BMI. Furthermore, associated obesity, such as Blautia obeum Ruminococcus torques, played pivotal roles regulating these activities. In conclusion, intervention may assist by modulating functions distasonis, obeum, torques could be targets for microbiota-based interventions.
