ABSTRACT Inflammation‐associated diseases have become widespread and pose a significant threat to human health, the therapeutic methods for diverse are inadequate due undesirable effects of synthetic ingredients. Recently, more evidence indicated that phytochemicals, plant secondary metabolites, numerous functions against via affecting variety mechanisms with their distinct advantages high efficiency low toxicity. Here, we highlight phytochemicals hinder inflammation‐associated (including Inflammatory diseases, cardiovascular metabolic syndrome, neurological disorders, skin respiratory kidney gastrointestinal retinal viral infections) by regulating crosstalk among various signal cascades MicroRNAs, SIRT1, DNMTs, NF‐κB, NLRP3, TGF‐β, Gasdermin‐mediated pyroptosis pathway), which can be considered as novel potential strategy. Furthermore, could prevent virus infection disturbing different targets in replication cycle. However, natural plants shown limited bioavailability water solubility, use adjuvants such liposomal phytochemical nanoparticles phytochemicals‐phospholipid complex promote exhibit beneficial diseases. The purpose this review is explore molecular promising applications fields provide some direction.

Nonalcoholic fatty liver disease (NAFLD) is a growing global health concern that closely linked to metabolic syndrome, yet no approved pharmacological treatment exists. The Mediterranean diet (MD) emerged as first-line dietary intervention for NAFLD, offering and hepatoprotective benefits. Now conceptualized complex chemical matrix rich in bioactive compounds, the MD exerts antioxidant anti-inflammatory effects, improving insulin sensitivity lipid metabolism. Mitochondria play central role NAFLD pathophysiology, influencing energy metabolism, oxidative stress, homeostasis. Emerging evidence suggests MD's compounds enhance mitochondrial function by modulating phosphorylation, biogenesis, mitophagy. However, most research has focused on individual rather than whole, leaving gaps understanding its collective impact pattern. This narrative review explores how influence highlighting key pathways such substrate control, dynamics, efficiency. A literature search was conducted identify relevant studies MD, mitochondria, NAFLD. While promising, our remains incomplete, particularly when current knowledge limited lack of mechanistic comprehensive holistic impact. Future integrating cutting-edge experimental approaches needed elucidate intricate diet-mitochondria interactions. deeper influences essential developing precision-targeted nutritional strategies can effectively prevent manage disease.

Non-alcoholic fatty liver disease (NAFLD) is a challenging to interfere with and represents potential long-term risk factor for hepatic fibrosis cancer. The Xiezhuo Tiaozhi (XZTZ) formula, water extract from crude herbs, has been widely used as an anti-NAFLD agent through clinical observation. However, the underlying pharmacological mechanisms of XZTZ formula its impact on pathways against NAFLD have not elucidated. Our study aims investigate effects regulatory treat NAFLD. possible active components were identified using ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS) molecular docking. To further explore mechanisms, forty-eight 6-week-old male C57BL/6J mice given individual attention high-fat high-sugar diet (HFHSD) or relevant control (Ctrl) diets 16 weeks successfully construct mouse model. Subsequently, levels serum biochemicals, pathological changes in liver, pyroptosis assessed therapeutic formula. Further, LPS-induced RAW264.7 cells Immortalized Mouse Kupffer (ImKC) verify vitro. We 7 chemical compounds 2 targets plausible points treatment Subsequent histopathological analysis revealed marked steatosis lipid accumulation HFHSD while conditions effectively ameliorated by administration Additionally, our work demonstrated that could attenuate M1 polarization, promote M2 suppress via SIRT1 pathway tissue samples. Moreover, validation performed ImKC showing silencing weaken relative affirmed role was associated macrophage. These findings suggest alleviated mice. ameliorations are involving attenuation promotion anti-pyroptosis pathway.

Abstract: Pyroptosis is a novel pro-inflammatory mode of programmed cell death that differs from ferroptosis, necrosis, and apoptosis in terms its onset regulatory mechanisms. dependent on cysteine aspartate protein hydrolase (caspase)-mediated activation GSDMD, NLRP3, the release cytokines, interleukin-1 (IL-1β), interleukin-18 (IL-18), ultimately leading to death. Non-coding RNA (ncRNA) type does not encode proteins gene transcription but plays an important role other post-transcriptional links. NcRNA mediates pyroptosis by regulating various related factors, which we termed signaling pathway. Previous researches have manifested closely development liver diseases, essential for injury, alcoholic fatty disease (ALD), non-alcoholic (NAFLD), fibrosis, cancer. In this review, attempt address ncRNA-mediated pathway above diseases their pathogenesis recent years, briefly outline TCM (Traditional Chinese Medicine) intervene modulating pyroptosis, will provide strategy find new therapeutic targets prevention treatment future. With the global epidemic trend of obesity, non-alcoholic fatty liver disease (NAFLD) has become a significant cause chronic disease, seriously affecting human health. Medium-chain triglycerides (MCT) with acid chain length varying between 6 and 10 carbon atoms (most sources from coconut palm kernel oils), which exhibited activities to improve lipid metabolism, prevent cardiovascular diseases, enhance immunity. However, efficacy differences potential mechanisms MCT traditional long-chain vegetable oils (palm oil, PA; high oleic peanut OA) in obesity-induced NAFLD were still unclear. The present study treated mice different dietary lipids for 16 weeks. results showed that supplements significantly improved abnormal elevation weight gain blood reduced hepatic accumulation greater extent than PA OA. Furthermore, bile profiling indicated changed composition acids liver, concentrations cholic (CA), deoxycholic (DCA), β-muricholic (β-MCA), ursodeoxycholic (UDCA) increased chenodeoxycholic Acid (CDCA), taurochenodeoxycholic (TCDCA), hyodeoxycholic (HDCA), taurohyodeoxycholic (THDCA). Mechanistically, supplement upregulated FXR signal inhibited expression key genes triglyceride synthesis thereby reducing accumulation. In summary, exerted superior effect on OA improving NAFLD. These provided new evidence application treating

Low-dose radiation has been extensively employed in clinical practice, including tumor immunotherapy, chronic inflammation treatment and nidus screening. However, the damage on spleen caused by low-dose significantly increases risk of late infection-related mortality, there is currently no corresponding protective strategy. In present study, a novel compound preparation named CB001 mainly constituted Acanthopanax senticosus (AS) Oldenlandia diffusa (OD) was developed to alleviate splenic injury fractionated exposures. As our results show that, white pulp atrophy excessive apoptosis tissue induced exposure were ameliorated CB001. Mechanistically, BAX-caspase-3 signaling nucleotide-binding domain leucine-rich-repeat-containing family pyrin 3 (NLRP3) inflammasome demonstrated be involved radio-protective activity with selective activators. Furthermore, crosstalk between NLRP3 mediating clarified, which pro-apoptotic protein BAX but not antiapoptotic Bcl2 found downstream NLRP3. Our study that use drug product can potentially facilitate alleviation radiation-induced for patients receiving medical imaging diagnosis or therapy.
