Background Under hyperglycemic conditions, impaired intestinal barrier integrity leads to heightened level of inflammation, playing important roles in driving diabetic complications. Emerging evidence supports the implications neutrophil extracellular traps (NETs) pathogenesis diabetes. However, whether NETs contribute hyperglycemia-linked impairment remains be investigated. Moreover, baicalin, major chemical component Scutellaria baicalensis Georgi, is equipped with twofold protective and suppressive activities. Yet, it unclear if baicalin effective at mitigating NETs-mediated impairment. Methods To directly address mechanistic epithelial impairment, impact DNase I treatment or Padi4 gene deficiency on was first examined streptozotocin (STZ)-induced mice vivo . Next, pharmacological formation investigated high glucose- and/or lipopolysaccharides (LPS)-stimulated neutrophils vitro STZ-induced , respectively. Results The experiments confirmed that glucose LPS induced formation. viability tight junction cells. histological immunohistochemical examinations unveiled along morphology, citrullinated histone H3 (H3Cit), a marker NETs, specific Ly6G were readily detected epithelium mice. Without affecting presence neutrophils, markedly mitigated improved morphology Notably, suppressed inhibited citrullination stimulated by glucose, both Furthermore, blunted partially preserved Conclusions current study sheds new light pathophysiological under conditions. Most importantly, findings here demonstrate for time inhibits LPS, which may part account its effects protecting against

Flavonoids are a class of important polyphenolic compounds, renowned for their antioxidant properties. However, recent studies have uncovered an additional function these natural flavonoids: ability to inhibit ferroptosis. Ferroptosis is key mechanism driving cell death in central nervous system (CNS) diseases, including both acute injuries and chronic neurodegenerative disorders, characterized by iron overload-induced lipid peroxidation dysfunction the defense system. This review discusses therapeutic potential flavonoids from herbs nutraceuticals as ferroptosis inhibitors CNS focusing on molecular mechanisms, summarizing findings preclinical animal models, providing insights clinical translation. We specifically highlight such Baicalin, Baicalein, Chrysin, Vitexin, Galangin, Quercetin, Isoquercetin, Eriodictyol, Proanthocyanidin, (−)-epigallocatechin-3-gallate, Dihydromyricetin, Soybean Isoflavones, Calycosin, Icariside II, Safflower Yellow, which shown promising results models injuries, ischemic stroke, cerebral ischemia-reperfusion injury, intracerebral hemorrhage, subarachnoid traumatic brain spinal cord injury. Among these, Baicalin its precursor Baicalein stand out due extensive research favorable outcomes injury models. Mechanistically, not only regulate Nrf2/ARE pathway activate GPX4/GSH-related pathways but also modulate metabolism proteins, thereby alleviating overload inhibiting While show promise especially settings, further needed evaluate efficacy, safety, pharmacokinetics, blood-brain barrier penetration application.

Flavonoids are known for their antioxidant, anti-inflammatory, antitumoral, and antiviral properties, as is the case baicalein derived from roots of Scutellaria baicalensis, which effective against respiratory viral infections. In this study, we investigate molecular mechanisms underlying interaction between Langmuir monolayers models cell membranes. For comparison, analyzed lipid extracts two lines: oropharyngeal carcinoma (HEp-2), susceptible to infections, primary melanoma (A375), not. Baicalein incorporation into A375 extract shifted π-A isotherms larger areas, reducing monolayer stability. contrast, its HEp-2 smaller enhancing both compaction Polarization-modulation infrared reflection-absorption spectroscopy (PM-IRRAS) revealed that interactions with involved electrostatic attractions repulsions choline phosphate headgroups, disrupting chain organization expanding monolayer. extracts, interacted strongly headgroups chains, increasing order stabilizing These findings suggest stabilizes membranes, potentially providing a protective mechanism Its selective membranes consistent therapeutic potential role in modulating membrane properties inhibit entry.
