Abstract Pain is estimated to affect more than 20% of the global population, imposing incalculable health and economic burdens. Effective pain management crucial for individuals suffering from pain. However, current methods assessment treatment fall short clinical needs. Benefiting advances in neuroscience biotechnology, neuronal circuits molecular mechanisms critically involved modulation have been elucidated. These research achievements incited progress identifying new diagnostic therapeutic targets. In this review, we first introduce fundamental knowledge about pain, setting stage subsequent contents. The review next delves into underlying disorders, including gene mutation, epigenetic modification, posttranslational inflammasome, signaling pathways microbiota. To better present a comprehensive view research, two prominent issues, sexual dimorphism comorbidities, are discussed detail based on findings. status quo evaluation manipulation summarized. A series improved innovative strategies, such as therapy, monoclonal antibody, brain-computer interface microbial intervention, making strides towards application. We highlight existing limitations future directions enhancing quality preclinical research. Efforts decipher complexities pathology will be instrumental translating scientific discoveries practice, thereby improving bench bedside.

Transcutaneous vagus nerve stimulation (tVNS) is an emerging non-invasive technique designed to stimulate branches of the distributed over body surface. Studies suggest a correlation between brain-gut-microbiota (BGM) axis and pathogenesis Alzheimer’s disease (AD). The BGM represents complex bidirectional communication system, with being crucial component. Therefore, electrical might have potential modify—most time probably in non-physiological way—the signal transmission within axis, potentially influencing progression or symptoms AD. This review explores interaction percutaneous emphasizing its effects on It examines various aspects, such as specific brain regions, gut microbiota composition, maintenance intestinal environmental homeostasis, inflammatory responses, plasticity, hypothalamic–pituitary–adrenal (HPA) regulation. suggests that tVNS could serve effective strategy modulate intervene treatment future.

Depression frequently occurs in patients with liver cirrhosis, yet the reasons for this correlation are not fully understood. Dysbiosis of gut microbiota has been implicated depression through gut–brain axis via vagus nerve. This study explored potential role gut–liver–brain nerve depression-like phenotypes mice cirrhosis. These underwent common bile duct ligation (CBDL), a method used to stimulate To assess behaviors, behavioral tests were conducted 10 days following either sham or CBDL surgeries. The displayed symptoms such as splenomegaly, elevated plasma levels interleukin-6 and tumor necrosis factor-α, decreased synaptic proteins prefrontal cortex (PFC), disrupted balance, changes blood metabolites (or lipids). Additionally, there positive negative correlations between relative abundance microbiome data Significantly, these reversed by performing subdiaphragmatic vagotomy. Intriguingly, improved after single injection arketamine, new antidepressant. results suggest that CBDL-induced mediated nerve, arketamine might offer treatment approach cirrhosis patients.

3,4-Methylenedioxymethamphetamine (MDMA) is the most widely used illicit substance worldwide. Nevertheless, recent observational studies demonstrated that lifetime MDMA use among U.S. adults was associated with a lower risk of depression and suicide thoughts. We recently reported gut-brain axis may contribute to MDMA-induced stress resilience in mice. To further explore this, we investigated effects subdiaphragmatic vagotomy (SDV) modulating mice subjected chronic restrain (CRS). Pretreatment (10 mg/kg/day for 14 days) blocked anhedonia-like behavior reduced expression synaptic proteins brain-derived neurotrophic factor prefrontal cortex (PFC) CRS-exposed Interestingly, SDV beneficial on these alterations Analysis gut microbiome revealed four measures α-diversity between sham + CRS group group. Moreover, specific microbes differed vehicle group, differences microbial composition were observed all groups. Untargeted metabolomics analysis showed prevented increase plasma levels three compounds [lactic acid, 1-(2-hydroxyethyl)-2,2,6-tetramethyl-4-piperidinol, 8-acetyl-7-hydroxyvumaline] positive correlations found two abundance several across In conclusion, our data suggest via vagus nerve might MDMA.

Many studies have shown that postoperative pain aggravates perioperative neurocognitive disorder (PND). In this study we aimed to clarify the effect of preoperative inflammatory on and cognitive function. We established model by injected complete freund adjuvant (CFA) PND tibial fracture surgery in 14- month-old C57BL/6 mice. The paw withdrawal threshold body weight mice were measured 7 days before 3 after surgery. On third day, subjected behavioral testing or sacrificed collect brain tissue. result shows CFA exacerbated dysfunction mice, enhanced surgery-induced activation microglia astrocytes hippocampus, increased overexpression IL-1β, IL-6, TNF-α, as well aggravated decreased expression α7nAChR HMGB1 hippocampus induced Our further aged rats, mechanism may be related neuroinflammation caused α7nAChR-mediated CAP high release HMGB1.

Abstract Over the past 2 decades, microbiome has received increasing attention for role that it plays in health and disease. Historically, gut was of particular interest to pain scientists studying nociplastic visceral conditions given anatomical juxtaposition these microorganisms neuroimmune networks drive such diseases. More recently, microbiomes both inside across surface body have been recognized driving sensory symptoms a broader set Microbiomes never more popular topic research, but date, there not systematic review preclinical literature. In this article, we identified all animal studies which manipulated behaviors were measured. Our analysis included 303 unique experiments 97 articles. Microbiome manipulation methods behavioral outcomes recorded each experiment so field-wide trends could be quantified reported. This specifically details species, injury models, behavior measures, manipulations used research. From analysis, also able conclude how alter thresholds naïve animals persistent intensity duration cutaneous models. summarizes by identifying existing gaps literature providing recommendations best plan, implement, interpret data collected experiments.
