Alzheimer’s Disease (AD), a progressive and debilitating condition, is reported to be the most common type of dementia, with at least 55 million people believed currently affected. Many causation hypotheses AD exist, yet intriguing link between viral infection its possible contribution known etiology has become an attractive focal point research for field challenging study task. In this review, we will explore historical perspective milestones that led investigate connection AD. Specifically, several viruses such as Herpes Simplex Virus 1 (HSV-1), Zika virus (ZIKV), severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), along others mentioned, include various presently considered within field. We delve into strong evidence implicating these in development also extend beyond mere associations by carefully analyzing potential mechanisms which may contribute pathology. This includes but not limited direct neuronal infections, dysregulation immune responses, impact on protein processing. Controversies challenges viral-AD relationship emerge tease out considered. Looking forward, emphasize future directions should take tackle remaining unanswered questions glaring gaps persist. Overall, review aims provide comprehensive survey past, present, infections their association development.

This review explores the therapeutic potential of intranasal chlorpheniramine maleate (iCPM) in managing both acute COVID-19 and Long COVID by integrating histamine H1 receptor antagonism bitter taste (T2R) activation. Current literature on histamine-mediated inflammation, T2R activation, dual-action mechanisms iCPM were analyzed. Emphasis was placed its antiviral, anti-inflammatory, mucosal immunity-enhancing properties. demonstrates significant efficacy addressing symptoms inhibiting inflammatory pathways reducing cytokine storms. As a agonist, it enhances immunity through nitric oxide production, mucociliary clearance, antimicrobial peptide synthesis, viral replication supporting respiratory health. Additionally, shows promise mitigating persistent long COVID, including fatigue, brain fog, dysfunction, chronic inflammation residual activity. The integration activation positions as novel dual-target therapy for infections. Its localized delivery broad mechanism action make promising candidate phases COVID-19. Future research should focus large-scale clinical trials personalized approaches based genetic variations pathways.

In this article we discuss the concept of accessible plasma membrane cholesterol and its involvement as a signalling molecule. Changes in cholesterol, although only being minor context total cell are key regulator overall cellular homeostasis by SREBP pathway. Accessible also provides second messenger between patched 1 smoothened hedgehog pathway important during development, depletion may provide mechanism resistance to microbial pathogens including SARS-CoV-2. We revise hypothesis that oxysterols form instance rapidly acting paracrine version cholesterol.

Oxysterols, or cholesterol oxidation products, are naturally occurring lipids which regulate the physiology of cells, including those immune system. In contrast to effects that mediated through nuclear receptors by epigenetic mechanism, take tens minutes occur, changes in activities cell-surface caused oxysterols can be extremely rapid, often taking place within subsecond timescales. Such receptor allow for regulation fast cellular processes, such as motility, secretion and endocytosis. These processes play critical roles both innate adaptive systems. This review will survey two broad classes (G-protein coupled (GPCRs) ion channels), mechanisms products act on them, their presence functions different cell types Overall, this highlight potential oxysterols, synthetic derivatives physiological therapeutic modulation

Post-COVID-19 syndrome (PCS) remains a major health issue worldwide, while its pathophysiology is still poorly understood. Systemic oxidative stress (OS) may be involved in PCS, which reflected by lower circulating free thiols (R-SH, sulfhydryl groups), as they are receptive to rapid oxidation reactive species. This study aimed investigate the longitudinal dynamics of serum R-SH after SARS-CoV-2 infection and association with development PCS individuals mild COVID-19.

Abstract Cholesterol plays essential roles in biological processes, including cell membrane stability and myelin formation. can be metabolized to oxysterols by enzymatic or nonenzymatic ways. Nonenzymatic cholesterol metabolites, also called cholesterol‐autoxidation are formed dependent on the oxidation of reactive oxygen species (ROS) such as OH• nitrogen species, ONOO − . Cholesterol‐autoxidation metabolites abundantly produced diseases inflammatory bowel disease atherosclerosis, which associated with oxidative stress. Recent studies have shown that further regulate immune system. Here, we review literature summarize how 25‐hydroxycholesterol (25‐OHC), 7α/β‐OHC, 7‐ketocholesterol, deal occurrence development infectious through pattern recognition receptors, inflammasomes, ROS production, nuclear G‐protein‐coupled receptor 183, lipid availability.
