ABSTRACT Sjogren’s Disease (SjD) is an autoimmune disorder characterized by salivary and lacrimal gland dysfunction immune cell infiltration leading to inflammation destruction. Although SjD a common disease, its pathogenesis not fully understood. In this study, we conducted single-cell transcriptome analysis of peripheral blood mononuclear cells (PBMC) from patients with symptomatic non-SjD controls identify types functional changes involved in pathogenesis. All PBMC populations showed marked differences gene expression between controls, particularly increase interferon (IFN) signaling signatures. T B displayed depletion ribosomal pathways linked protein translation. had increased frequencies naive cells, which featured unique profile (GEP) distinct hallmarks hyperactivation. Non-negative matrix factorization (NMF) also identified several non-overlapping GEPs CD4 + CD8 differential usage controls. Of these, only the Th1 activation GEP was enriched whereas other two were depleted emphasizing important role SjD.
