Abstract Organoid models have advanced our understanding of adult stem/progenitor cell dynamics and function. However, uncovering the regulatory mechanisms scarce often quiescent stem cells in organs like salivary glands remains challenging. Using single-cell RNA sequencing bulk ATAC RNA-sequencing analysis, we conducted in-depth profiling cellular populations key signaling pathways characterizing a mouse submandibular gland organoid (mSGO) model at different temporal stages response to radiation damage. The lacrimal gland (LG) is the major source of aqueous tears, and insufficient LG secretion leads to aqueous-deficient dry eye (ADDE) disease. To provide a foundational description LG's protein expression patterns, we prepared extracts LGs from wild-type an ADDE mouse model analyzed proteome by quantitative mass spectrometry. were isolated model, male non-obese diabetic (NOD) mice control BALB/c (n = 6 each). Background and Aim: The Indonesian red claw crayfish (Cherax quadricarinatus) represents a significant aquaculture commodity with substantial economic importance; however, disease outbreaks, particularly tail ulceration caused by Aeromonas hydrophila, pose challenges to sustainable production. This study evaluated the efficacy of fermented palm kernel meal (FPKM), rich in mannan oligosaccharides, as prebiotic additive enhance non-specific immune responses C. quadricarinatus. Materials Methods: A total 120 (mean weight 10.3 ± 0.15 g length 7.53 0.26 cm) were allocated completely randomized design comprising four dietary treatments: control group without FPKM probiotics three experimental groups receiving diets supplemented at concentrations 40, 80, g/kg along probiotics. Hemolymph samples collected baseline (day 0) on days 3, 5, 7, 14 post-treatment. Non-specific included hemocyte count (THC), differential (DHC), phagocytosis activity (PA), index (PI), phenol oxidase (PO) activity, superoxide dismutase (SOD) plasma protein (TPP), gene expression (lipopolysaccharide β-1,3-glucan-binding [LGBP], lectin). Results: inclusion significantly increased THC, peak enhancement observed day 7 highest concentration (120 g/kg). DHC remained stable across treatments. Summary Granulocyte-macrophage colony-stimulating factor (GM-CSF) has a non-redundant role in the emergence and maintenance of alveolar macrophages (AMs). However, its developmental steady-state myelopoiesis outside lung is largely unexplored. Scanning through developing tissues using Fate-map reporter GM-CSF mouse strain, we discovered that was produced by type 2 innate lymphoid cells (ILC2s) submandibular sublingual salivary gland (SG) during postnatal development. producing ILC2s foster development hitherto undescribed phagocyte subset, which named adenophages. Detailed analysis focusing on phenotypic transcriptional profiling revealed adenophages display shared aspects both, dendritic (DCs). We found them to be homogenously distributed across SG, but always close proximity myoepithelial cells. Importantly, were present throughout all analyzed exocrine glands such as lacrimal mammary glands, also identified human SG sections, indicating conserved species.
