Members of the claudin protein family are major constituents tight junction strands and determine permeability properties paracellular pathway. Novel transplantable organs need to be developed address the global organ shortage. Transplantation of embryonic kidney tissue, or metanephros, facilitates glomerular and tubular maturation offers partial functional support. However, adult environments do not permit exponential growth in size, limiting life-supporting functionality replacement effect this approach. Here, we a novel strategy that combines fusion bladders with multiple anastomoses host ureter, enabling significant increase metanephros transplantation urinary tract integration. By surgically anastomosing divided bladder segments, reconstructed excretory pathways by merging four metanephroi into each integrating them ureter. Following integration 20 at para-aortic region, anephric rats survived for over month generated approximately 50,000 nephrons vivo. Ultrastructural single-cell- transcriptomic analyses revealed maturity transplanted was comparable kidneys, although their small size likely contributed decreased urine concentration ability. Postoperative support helped normalize physiological homeostasis, including solute clearance, acid-base balance, electrolyte levels, hormone within vital ranges. Aging leads to functional decline across tissues, often accompanied by profound changes in cellular composition and cell-intrinsic molecular states. However, a comprehensive catalog of how the population individual cell types change with age associated epigenomic dynamics is lacking. Here, we constructed single-cell chromatin accessibility atlas consisting ∼7 million cells from 21 tissue spanning three groups both sexes. This dataset revealed 536 main 1,828 finer-grained subtypes, defined unique landscapes at ∼1.3 cis-regulatory elements. We observed widespread remodeling immune lineages, increases plasma macrophages, depletion T B progenitors. Additionally, non-immune populations, including kidney podocytes, ovary granulosa cells, muscle tenocytes lung aerocytes, showed marked reductions age.
