Abstract Platinum‐based chemotherapeutic agents are widely used in the treatment of cancer. However, their effectiveness is limited by severe adverse reactions, drug resistance, and poor water solubility. This study focuses on synthesis characterization new water‐soluble cationic monofunctional platinum(II) complexes starting from [PtCl(η 1 ‐C 2 H 4 OEt)(phen)] ( , phen=1,10‐phenanthroline) precursor, specifically [Pt(NH 3 )(η OEt)(phen)]Cl ), [Pt(1‐hexyl‐1 ‐imidazole)(η ‐benzo[d]imidazole)(η which deviate traditional requirements for antitumor activity. These were evaluated cytotoxic effects comparison to cisplatin, using immortalized cervical adenocarcinoma cells (HeLa), human renal carcinoma (Caki‐1), normal (HK‐2). While complex showed minimal cell lines, demonstrated higher cytotoxicity than cisplatin. Cisplatin (CIS) stands as one of the most effective chemotherapy drugs currently available. Despite its anticancer properties, clinical application CIS is restricted due to nephrotoxicity. Our research aimed specify impact ketotifen fumarate (KET) against nephrotoxicity induced by in mice. Male NMRI mice were treated with KET (0.4, 0.8, and 1.6 mg/kg, ip) for seven days. On fourth day study, a single dose (13 was administered, sacrificed on eighth day. The results indicated that administration attenuated CIS-induced elevation BUN Cr serum, well renal KIM-1 levels. This improvement accompanied significant reduction kidney tissue damage, which supported histopathological examinations. Likewise, decrease ratio GSH GSSG antioxidant enzyme activities (CAT, SOD, GPx), increase lipid peroxidation marker (TBARS) reversed KET-treated ELISA revealed ameliorated levels TNF-α, IL-1β, IL-18. Abstract Chemotherapy (CT) is one of the flagship options for treatment cancers worldwide. It involves use cytotoxic anticancer agents to kill or inhibit proliferation cancer cells. However, despite its clinical efficacy, CT triggers side effect toxicities in several organs, which may impact patient’s quality life and outcomes. While toxicity consistent with non-ferroptotic mechanisms involving oxidative stress, inflammation, mitochondrial impairment other aberrant signalling leading apoptosis necroptosis, recent studies show that ferroptosis, a non-apoptotic, iron-dependent cell death pathway, also involved pathophysiology organ toxicity. provokes ferroptosis via system Xc – /GPX-4/GSH/SLC7A11 axis depletion, ferritinophagy, iron overload, lipid peroxidation upregulation ferritin-related proteins.
