Mitochondria
are
essential
double-membrane
organelles
with
intricate
structures
and
diverse
functions
within
cells.
Under
normal
physiological
conditions,
mitochondria
regulate
cellular
metabolism
maintain
energy
homeostasis
via
the
electron
transport
chain,
mediate
stem
cell
fate,
modulate
reactive
oxygen
species
production,
playing
a
pivotal
role
in
supply
lifespan
extension.
However,
mitochondrial
dysfunction
can
lead
to
various
pathological
changes,
including
aging,
necrosis,
dysregulated
tumor
immunity,
initiation
progression
of
cancer.
Moreover,
abnormal
is
closely
associated
numerous
diseases,
such
as
neurodegenerative
disorders,
metabolic
syndromes,
cancers.
In
recent
years,
targeting
has
emerged
promising
anticancer
strategy,
aiming
for
therapeutic
benefits.
Nonetheless,
approaches
face
limitations,
low
delivery
efficiency
insufficient
specificity.
This
review
systematically
explores
structure
function,
their
roles,
potential
challenges
mitochondria-targeted
strategies
cancer
therapy,
providing
insights
future
research
directions.

A
century
ago,
the
Warburg
effect
was
first
proposed,
revealing
that
cancer
cells
predominantly
rely
on
glycolysis
during
process
of
tumorigenesis,
even
in

oxygen,
shifting
main

metabolism
from
tricarboxylic
acid
cycle
to
aerobic
glycolysis.
Recent
studies
have
unveiled
dynamic
transfer
mitochondria
within
tumor
microenvironment,

cells,
immune

In
this
review,
we
explore
pathways

these

promote
aggressiveness,
