The Flaviviridae family includes the dengue virus (DENV). About half of world's population is in danger because estimated 390 million infections and 96 symptomatic cases that occur each year. An effective treatment for fever (DF) does not yet exist. Therefore, a better knowledge how viral proteins virus-targeted medicines may exert distinct functions depending on exact cellular region addressed aid creating much-needed antiviral medications. Lipids facilitate coordination many replication phases, from entrance to dissemination. In addition, flaviviruses masterfully plan significant rearrangement host cell's lipid metabolism foster growth new viruses. Recent research has consistently shown significance certain classes flavivirus infections. For instance, DENV-infected cells, overall cholesterol (CHO) levels are only little altered, DENV significantly reduced when CHO inhibited. Moreover, statins decrease serotype 2 (DENV-2) titers, indicating prerequisite cycle. Furthermore, Proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors now being evaluated human research. A pharmacological target management high PCSK9. suppression PCSK9 been proposed as possible defense against DENV. Numerous studies have generally recommended use lipid-lowering medications suppress As result, we investigated popular techniques this We also examined metabolism, lipids, receptors affect regulation. Human immunodeficiency virus (HIV) and hepatitis B (HBV) continue to be global public health issues. Globally, about 39.9 million persons live with HIV in 2023, according the Joint United Nations Programme on HIV/AIDS (UNAIDS) 2024 Fact Sheet. Consequently, World Health Organisation (WHO) reported that 1.5 new cases of HBV occur, approximately 820 thousand mortalities yearly. Conversely, lower percentage (30%) receive a diagnosis is setback achieving WHO 2030 target for zero globally. This has necessitated concern repurpose antiretroviral (ARV) drugs treatment diseases. review provides an introductory background, including pros cons repurposing antiretrovirals (ARVs) treatment. We examine similarities replication mechanisms between HBV. further investigate some clinical studies trials co-infected mono-infected patients HIV–HBV. The topical keywords ARV drugs, therapy, Hepatitis title, abstracts are searched PubMed, Web Science, Google Scholar. advanced search includes period 2014–2024, full text, trials, randomized control review. results filtered from 361 51 relevant articles. investigations revealed replicate via common route known as ‘reverse transcription’. Clinical trial indicate early initiation ARVs, particularly tenofovir disoproxil fumarate (TDF) part regimen, significantly reduced viral load patients. In HBV, timely correct precise medication essential reduction. Therefore, genetic profiling pivotal successful drug Pharmacogenetics enables prediction right dosages, specific individual responses, reactions. study uniquely explores intersection pharmacogenetics optimized therapy. Additional vivo, silico research important validation potency, optimum dosage, safety repurposed Furthermore, prioritization collaborations comprising regulators funders foster clinically adopting incorporating ARVs therapy recommended.

Rubella infection (RuV) during early pregnancy is a known cause of congenital rubella syndrome (CRS). However, the mechanisms by which virus crosses placenta and infects fetus are not fully understood. It has been that various kinds cell stresses can occur formation. Previously, we demonstrated low-glucose-induced endoplasmic reticulum stress could drastically enhance RuV in immortalized human first-trimester trophoblast cells. In this study, investigated roles oxidative these Oxidative was induced Swan.71 cells culturing them medium containing hydrogen peroxide (H2O2) concentrations durations (50 µM or 100 for 24 h, 150 1 h). with clinical strain performed h post-treatment, capsid proteins were visualized at 48 post-infection (hpi) using flow cytometry (FCM) fluorescence microscopy (IF), respectively. The findings significantly enhanced infection, as evidenced FCM analysis, showing twofold increase rate, confirmed IF assay. Additionally, increased intracellular viral replication observed 3 dpi. These suggest may promote maternal-to-fetal transmission rubella, contributing to development CRS.

Severe fever with thrombocytopenia syndrome (SFTS) is a rapidly progressing infectious disease caused by novel bunyavirus characterized high fever, thrombocytopenia, and multiple organ damage. While lipids play an important role in viral infections, the specific alterations lipid metabolism during SFTSV infection remain unclear. This study aimed to elucidate global metabolic profiles of SFTS patients mild, severe, fatal outcomes. A total 60 patients, consisting 30 15 severe healthy controls, were enrolled for investigation lipidomics serum using liquid chromatography-tandem mass spectrometry (LC-MS/MS). Our findings revealed signature induced further confirmed that glycerophospholipid pathway was profoundly affected progression outcomes patients. Importantly, LysoPC (20:0) (P-16:0) are strongly correlated clinical parameters infection. Furthermore, we demonstrated substantial prognostic value receiver operating characteristic (ROC) curve analysis, providing evidence their remarkable as biomarkers predicting In particular, (P-16:0), along APTT, yielded superior performance [area under (AUC) = 98.4%], outperforming routine parameters. Collectively, our lipidomic landscape after infection, which offers new insights into mechanisms suggests targeting may serve potential therapeutic strategy.
