Multidrug efflux pumps are critical elements in both intrinsic and acquired antibiotic resistance of bacterial populations. Consequently, most studies regarding these protein machineries focus on this specific phenotype. Nevertheless, different works show that participate other aspects physiology too. Herein, we study the Pseudomonas aeruginosa multidrug pump MexJK. Previous studies, using model strains lacking MexAB-OprM MexCD-OprJ pumps, support MexJK can extrude erythromycin, tetracycline, triclosan. However, results here reported indicate potential increased extrusion, a mutant overexpressing mexJK, does not alter antibiotics susceptibility wild-type genetic background where all remain functional. clear impact quorum sensing (QS) response, mainly Pqs-dependent QS regulation network expression Pqs-regulated virulence factors, was observed linked to mexJK overexpression. The production siderophore pyoverdine strongly depended level expression, suggesting might P. pyoverdine-dependent iron homeostasis. All all, presented current article functions as MexJK, go beyond modulate relevant physiology.

Introduction The resistance-nodulation-division (RND) family is the most important group of multidrug efflux pumps in Gram-negative bacteria. Their inhibition increases susceptibility these microorganisms to antibiotics. study effect pumps' overexpression on bacterial physiology antibiotic-resistant mutants allows for identification exploitable weaknesses associated with resistance acquisition.Areas covered authors describe different RND strategies and provide examples inhibitors. This review also discusses inducers expression pumps, used human therapy that can produce transient antibiotics vivo. Since may have a role virulence, use systems as targets search antivirulence compounds discussed. Finally, this analyzes how trade-offs acquisition mediated by guide tackle such resistance.Expert opinion Increasing knowledge regulation, structure function provides information rational design pump These inhibitors would increase several and, occasionally, will reduce virulence. Furthermore, has serve develop new anti-resistance strategies.

Pseudomonas aeruginosa is a pathogen that causes healthcare-associated infections (HAIs) worldwide. It unclear whether P. isolated from the natural environment has same pathogenicity and antimicrobial resistance potential as clinical strains. In this study, virulence- resistance-associated genes were compared in 14 genomic sequences of environmental isolates using VFDB, PATRIC, CARD databases. All found to share 62% virulence related adhesion, motility, secretion systems, quorum sensing 72.9% efflux pumps membrane permeability. Our results indicate both types possess conserved genetic information associated with mechanisms regardless source. However, none high-risk clones (HRCs). These (ST235 ST111) only isolates, which have an impact on human medical epidemiology due their ability spread persist, indicating correlation between increased virulence. The variation antibiotic susceptibility suggest biotechnological applications if obtained sources are under surveillance investigation limit emergence resistant strains

The first total synthesis of the repeating units O-antigens Pseudomonas aeruginosa ATCC 27577, O10, and O19 was achieved via a linear glycosylation strategy. This also represents an oligosaccharide containing α-linked N-acetyl-l-galactosaminuronic acid (l-GalpNAcA) unit. All glycosyl linkages, including three challenging 1,2-cis-glycosidic bonds amino sugars, were effectively constructed with high to exclusive stereoselectivity, while orthogonal protection tactics employed facilitate regioselective glycosylations introduction variety functionalities. An acetyl group migration phenomenon found during O-acylated unit P. 27577 antigen. synthetic targets carried functional in linker at reducing end, thus facilitating further elaboration biological studies. strategy established here should be useful for preparation other similar oligosaccharides.

Background/Objectives: Pseudomonas aeruginosa causes chronic infections in humans and animals, especially cats dogs. This bacterium’s ability to adapt acquire antibiotic resistance traits may complicate exacerbate antibacterial therapy. study aimed evaluate the patterns, virulence factors form biofilms of P. strains isolated from Algerian dogs cats. Methods: Nineteen samples were collected healthy diseased Isolates studied for their antibiotic-resistance patterns (disc diffusion method) biofilm formation (Microtiter assay) whole-genome sequenced (MinION). Results: (15 4 cats) isolated. Antibiotic-resistance phenotypes observed against amoxicillin–clavulanic acid (100%); meanwhile, towards ticarcillin was 40% (dogs) 25% (cats), ticarcillin–clavulanic 13.33% cats, respectively, imipenem 75% (cats) 20% (dogs). Moreover, 95% biofilm-producers. Different antimicrobial genes (ARGs) found: beta-lactamase genes, mainly PAO, OXA-494, OXA-50 OXA-396 an aminoglycoside gene (aph(3′)-IIb). The main high-risk sequence types (STs) ST244, 2788, 388 1247. A large panel detected: exoS, exoT, exoY, lasA, toxA, prpL, algD, rhIA others. Conclusions: genetic variety resistant virulent makes public health protection difficult. Continuous monitoring research compliance with One Health policy are needed solve this problem.

This study analyzed eleven isolates of colistin-resistant Pseudomonas aeruginosa, originating from Portugal and Taiwan, which are associated with various pathologies. The results revealed significant genetic diversity among the isolates, each exhibiting a distinct profile. A prevalence sequence type ST235 was observed, characterizing it as high-risk clone, serotyping indicated predominance O11, chronic respiratory infections in cystic fibrosis (CF) patients. phylogenetic analysis demonstrated clades complex evolutionary relationships. Additionally, transposable elements such Tn3 IS6 were identified all highlighting their importance mobility antibiotic resistance genes. An antimicrobial profiles pan-drug high genes conferring to β-lactams aminoglycosides. Furthermore, additional analyses mutations regulatory networks specific loci previously implicated colistin resistance, pmrA, cprS, phoO, others, suggesting possible contribution observed resistant phenotype. has strong impact because not only reveals mechanisms P. aeruginosa but also identifies resistance.

Single-stranded DNA (ssDNA)-binding protein (SSB) plays a crucial role in replication, repair, and recombination as well replication fork restarts. SSB is essential for cell survival and, thus, an attractive target potential antipathogen chemotherapy. Whether naturally occurring products can inhibit remains unknown. In this study, the effect of flavonols myricetin, quercetin, kaempferol, galangin on inhibition Pseudomonas aeruginosa (PaSSB) was investigated. Furthermore, identified novel quercetin-binding protein. Through electrophoretic mobility shift analysis, myricetin could ssDNA binding activity PaSSB with IC50 2.8 ± 0.4 μM. The insignificant. To elucidate flavonol specificity, crystal structure complexed non-inhibitor quercetin solved using molecular replacement method at resolution 2.3 Å (PDB entry 7VUM) compared inhibitor 5YUN). Although bound similar site, their poses were different. Compared aromatic ring shifted by distance 4.9 angle 31o hydrogen bonding to side chain Asn108 PaSSB. addition, occupied interacted sites Lys7 Glu80 whereas did not. This result might explain why could, but not, strongly evidence reveals specificity also extends interactomes natural anticancer include OB-fold SSB.
