Fibrous scaffolds capable of delivering natural drugs and herbs show great promise for tissue regeneration wound care, particularly in personalized medicine. This study presents the fabrication characterization drug-eluting antibacterial core–shell mats composed polycaprolactone (PCL) pectin nanofibers produced through coaxial electrospinning. Berberine chloride (BBR), an herbal compound with antineoplastic, anti-inflammatory, antilipidemic, antidiabetic properties, served as model drug. Poly(vinyl alcohol) (PVA) was blended to enhance mechanical properties core fibers. The shell modified two-dimensional Ti3C2Tx (MXene) nanosheets subjected covalent ionic cross-linking. Structural analysis confirmed successful production bead-free fibers diameters ranging from 160 350 nm, depending on composition. PCL were uniformly coated a pectin/PVA approximately 90 nm thick. inclusion BBR MXene increased fiber diameter. Drug-release kinetics, modeled by using Korsmeyer-Peppas, revealed two-stage release mechanism. Pseudomonas aeruginosa is a common gram-negative opportunistic pathogen that now commonly treated with carbapenems, such as Meropenem. However, the increasing rate of emergence heteroresistant strains poses therapeutic challenge. Therefore, we examined antibacterial activity Sodium houttuyfonate (SH, compound derived from Houttuynia cordata) in combination Meropenem (MEM) against and investigated mechanism houttuyfonate. Heteroresistant was used experimental strain for study combined action two drugs inves-tigated by determining Minimum Inhibitory Concentration (MIC), Fractional Index (FICI), time killing curves. Also effect on biofilm well bacterial swimming motility assay crystal violet staining biofilm, microanalysis assay, quantitative real-time PCR (qRT-PCR) population sensing related virulence factors. For screened strains, MIC SH 4,000 μg/ml; FICI both four ≤0.5, which showed synergistic effect. When ≥ 250 μg/ml, it able to effectively inhibit formation ability compared blank control group. In qRT-PCR experiment, expression formation-related genes (pslA, pelA, aglD, lasI, lasR, rhlA) ability-related (fliC, pilZ, pilA) were decreased SH-treated group, Our demonstrated exhibited inhibition aeruginosa, may achieve its inhibitory inhibiting formation, motility, down-regulating genes.

Background Carbapenem-resistant Klebsiella pneumoniae (CRKP) infections pose a major clinical challenge due to multidrug resistance. This study evaluated the features, antibiotic resistance mechanisms, virulence factors, and potential therapeutic impact of berberine hydrochloride (a traditional Chinese medicine) in CRKP infections. Methods Ninety-four isolates from bloodstream at First Affiliated Hospital Zhejiang Medical University were characterized for carbapenemase genes, susceptibility, determinants. Clinical data analyzed identify risk factors infection, vitro antibacterial activity was assessed. Results Most (71.3%) intensive care unit (ICU) patients. The bla KPC gene predominant mechanism (62.77%), while genes uge (93.62%) wabG (92.55%) highly prevalent.
