Object To clarify the clinical efficacy of washed microbiota transplantation (WMT) for metabolic syndrome (MetS), and explore differences in profile bacterial outer membrane vesicles (OMVs) donor fecal bacteria suspension received by MetS patients with good poor outcomes, to construct a predictive model WMT using differential metabolites. Methods Medical data 65 who had completed at least 2 courses from 2017.05 2023.07 were collected. Fecal donors collected, treated during this period collected as well. The changes BMI, blood glucose, lipids, pressure other indicators before after compared. OMVs isolated off-target metabolomic sequencing was performed Liquid Chromatograph Mass Spectrometer (LC–MS). Results Compared baseline, Body mass index (BMI), Systolic (SBP) Diastolic (DBP) showed significant decreases 1st (short-term) 2nd (medium-term) courses, fasting glucose (FBG) also session. There difference between Marked Response Moderate OMVs. It that 960 metabolites significantly up-regulated 439 down-regulated. ROC suggested 9-carboxymethoxymethylguanine, AUC = 0.8127, 95% CI [0.6885, 0.9369], most potent metabolite predicting available efficacy. Conclusion short-term medium-term MetS. structure level 9-Carboxy methoxy methylguanine can be predictor treatment

A variety of hypoglycaemic drugs are used to treat polycystic ovarian syndrome (PCOS), but their efficacy remains insufficient. Glucokinase activators (GKAs) a unique class medications with emerging potential, notably in significantly reducing insulin resistance (IR). Nevertheless, the GKAs treating PCOS, particularly absence or presence IR, uncertain. The meta-analysis protocol aims address this knowledge gap, furnish evidence-based data support potential revisions PCOS treatment guidelines and promote utilisation clinical settings. comprehensive search will be conducted across Cochrane Central Register Controlled Trials, PubMed, Web Science, Embase, Medline, Scopus, CNKI, Wanfang VIP databases identify randomised controlled trials investigating use irrespective IR. encompass all available studies without language restrictions cover period from inception each database 10 April 2024. Disputes resolved by talking third expert following screening articles extraction two reviewers. primary outcomes interest changes anthropometric parameters, menstrual frequency, sex hormone levels, glucose metabolism, while secondary objectives include lipid metabolism adverse events. methodological quality study assessed using Version 2 Collaboration tool for assessing Risk Bias (RoB 2.0), Grade Recommendations, Assessment, Development Evaluation (GRADE) technique assess evidence degree recommendation. duration 5 2024 2025. Since just analyses that readily public does not directly involve patient participation, ethical approval is necessary. findings made being published medical journal subject peer review.
