In this study, we investigated the behavior of extracellular vesicles (EVs), during capillary isoelectric focusing (cIEF). For that, used different approaches, imaging cIEF with a whole-column detection (WCID) and conventional as well methods (LIF after EV labelling, native fluorescence UV). Our study reveals that EVs exhibit significant aggregation their migration toward, upon reaching, point (pI). By optimizing key parameters such voltage addition solubilizers, successfully reduced issue, particularly bovine milk EVs. findings also showed distinct pI regions observed for isolated from sources: shows acidic characteristics (4.0-4.1), while pig human plasma more basic zones (4.7-4.9 5.8-6.7, respectively). The was extended to coupled laser induced (LIF) using intra-vesicular CFDA-labeled EVs, better understand susceptibilities. Prolonged mobilization time due long lengths adversely affected EV's integrity in cIEF. necessity specific optimization each source variations charge distribution across regions. use short length (<10 cm), low electric field solubilizers Tween-20 is recommended preserve cIEF-EV studies.
