The RIG-I-like receptors (RLRs), comprising retinoic acid-inducible gene I (RIG-I), melanoma differentiation-associated 5 (MDA5), and laboratory of genetics physiology 2 (LGP2), are pattern recognition belonging to the DExD/H-box RNA helicase family proteins. RLRs detect viral RNAs in cytoplasm respond by initiating a robust antiviral response that up-regulates interferon cytokine production. RIG-I MDA5 complement each other recognizing different features, LGP2 regulates their activation. RIG-I's multilayered proofreading mechanisms ensure accurate detection while averting harmful responses host RNAs. C-terminal domain targets 5'-triphosphate double-stranded (dsRNA) blunt ends, an intrinsic gating mechanism prevents domains from non-specifically engaging with ATPase translocation activity adds another layer selectivity minimizing lifetime on non-specific RNAs, preventing off-target versatility function also amplifies downstream signaling enhancing (CARDs) exposure dsRNA promoting oligomerization. Transposable elements (TEs) are of interest as immunomodulators for cancer therapies. TEs can fold into dsRNAs that trigger the interferon response. Here, we investigated effect different HDAC inhibitors (HDACIs) on expression in leiomyosarcoma cells. Our data show endogenous retroviruses (ERVs), especially ERV1 elements, upregulated after treatment with HDAC1/2/3-specific inhibitors. Surprisingly, response was not activated. We observed an increase A-to-I editing ERV1. This could have impact stability and activation also found H3K27ac levels increased LTR12 subfamilies, which be regulatory controlling proapoptotic genes such TNFRSF10B. In summary, provide a detailed characterization modulation to HDACIs suggest use combination ADAR induce cell death support immunotherapy cancer.
