Thioether oxidation to sulfoxide by H2O2 has been widely reported as an ROS-sensitive trigger in drug delivery applications. Through a number of straightforward kinetic experiments with series aryl thioethers, we show that under near-physiological conditions is expected have half-lives on the scale hundreds hours at pathophysiologically relevant concentrations. On other hand, hypochlorite can oxidize thioethers much faster rates range seconds and minutes sulfone similar conditions. Such information means likely plays more important role than activating thioether-based systems.
