Alzheimer's disease currently has no cure and is usually detected too late for interventions to be effective. In this study we have focused on cognitively normal subjects the impact of risk factors their long-range brain connections. To detect vulnerable connections, devised a multiscale, hierarchical method spatial clustering whole tractogram examined age APOE allelic variation cognitive abilities bundle properties including texture e.g., mean fractional anisotropy, variability, geometric streamline length, volume, shape, as well asymmetry. We found that third level subdivision in hierarchy provided most sensitive ability genotype differences associated with factors. Our results indicate frontal bundles were major predictor, while occipital cortex cerebellar connections important predictors heavily dependent, showed accelerated decline shape similarity, increased Cognitive metrics related olfactory memory mapped bundles, providing possible early markers neurodegeneration. addition, physiological such diastolic blood pressure changes white matter tracts. novel data driven analysis tractography may differentiate populations at AD isolate specific networks.
