Abstract Depression is a mood disorder that may increase risk for the development of insulin resistance (IR) and type 2 diabetes (T2D), vice versa. However, mechanistic pathway linking depression T2D not fully elucidated. This state-of-the-art review explores the relationship between depression and diabetes, highlighting two-way influences that make treatment challenging worsen outcomes of both conditions. Depression diabetes often co-occur share genetic, lifestyle, psychosocial risk factors. Lifestyle elements such as diet, physical activity, sleep patterns play a role on development management conditions, need for integrated strategies. The evidence suggests traditional strategies focusing either condition in isolation fall short addressing intertwined nature depression. Instead, care models encompassing psychological support medical are recommended to improve efficacy patient adherence. Such require collaboration across multiple healthcare disciplines, including endocrinology, psychiatry, primary care, offer holistic approach care. also identifies significant patient-related barriers effective management, stigma, resistance, health literacy, which be addressed through patient-centered education systems. Purpose of review The circular interactions between type 2 diabetes (TMD2) and major depressive disorder (MDD) are well documented but the understanding their mechanisms has only recently gained more clarity. Background Diabetic retinopathy (DR) is the most frequent complication of type 2 diabetes and remains leading cause preventable blindness. Current clinical decisions regarding administration antidiabetic drugs do not sufficiently incorporate risk DR due to inconclusive evidence from preceding meta-analyses. The high comorbidity of major depressive disorder (MDD) with other diseases has been well-documented. However, the pairwise causal connections for MDD comorbid networks are poorly characterized. We performed Phenome-wide Mendelian randomization (MR) analyses to explore bidirectional associations between (N = 807,553) and 877 common from FinnGen datasets 377,277). inverse variance weighting method was primary technique, methods (weighted median MR-Egger) were used sensitivity analyses. Our MR showed that genetic liability is causally associated risks 324 disease phenotypes (average b: 0.339), including 46 psychiatric behavioral disorders 0.618), 18 neurological 0.348), 44 respiratory 0.345), 40 digestive 0.281), circulatory 0.237), 37 genitourinary 0.271), 66 musculoskeletal connective 0.326), 22 endocrine 0.302), others. For most analyses, little evidence heterogeneity pleiotropy detected. findings confirmed extensive significant role predisposition in contributing human phenotypes, which more pronounced than those seen analysis influences on MDD.

Abstract The co-occurrence of insulin resistance (IR)-related metabolic conditions with neuropsychiatric disorders is a complex public health challenge. Evidence the genetic links between these phenotypes emerging, but little currently known about genomic regions and biological functions that are involved. To address this, we performed Local Analysis [co]Variant Association (LAVA) using large-scale (N=9,725-933,970) genome-wide association studies (GWASs) results for three IR-related (type 2 diabetes mellitus, obesity, syndrome) nine disorders. Subsequently, positional expression quantitative trait locus (eQTL)-based gene mapping downstream functional analyses were on significant loci. Patterns negative positive local correlations (|r g |=0.21-1, p FDR <0.05) identified at 109 unique across all phenotype pairs. emerged even in absence global Alzheimer’s disease, bipolar disorder, Tourette’s syndrome. Genes mapped to correlated showed enrichment pathways integral immune-inflammatory function, vesicle trafficking, signalling, oxygen transport, lipid metabolism. Colocalisation further prioritised 10 genetically likely harbouring shared causal variants, displaying high deleterious or regulatory potential. These variants found within close proximity genes, such as SLC39A8 HLA-DRB1 , can be targeted by supplements already drugs, including omega-3/6 fatty acids, immunomodulatory, antihypertensive, cholesterol-lowering drugs. Overall, our findings underscore landscape IR-neuropsychiatric multimorbidity, advocating an integrated disease model offering novel insights research treatment strategies this domain. Highlights correlations. Both Enrichment immune, signalling pathways, among others. Pinpointed regions. Identified therapeutic targets, e.g., HLA-DRB1, drug repurposing.

Objectives: Comorbid diabetes is an important factor in the treatment of major psychiatric disorders. However, a substantial proportion diabetic patients remain undetected by routine indices such as blood glucose and HbA1c. This study tried to estimate rates unidentified or prediabetic using 75 g oral tolerance test (OGTT). Methods: Participants were 25 with depressive disorder (MDD), 28 bipolar (BP), 26 schizophrenia, psychiatrically normal controls. They all Japanese, showed non-diabetic levels (<126 mg/dL) HbA1c (<6.0%). Results: Relatively high mellites (DM)-type abnormality (32% MDD, 21.4% BP 42.3% schizophrenia v. 10.7% controls). The difference between control groups was statistically significant (p = 0.008). When abnormal metabolism defined state (either glycemia, impaired fast glycemia tolerance) DM type OGTT, it more frequently seen than controls (64% 46.4% 46.2% 35.7% Individuals higher values compared those < 0.001) 0.021) states. Conclusions: results suggest that relatively proportions for metabolism, indicating importance OGTT even if

Triglyceride glucose (TyG) index has been suggested as an alternative indicator of insulin resistance (IR); however, the association between TyG and suicide attempts (SA) in major depressive disorder (MDD) is unclear. The aim this study was to investigate relationship SA Chinese patients with first-episode drug-naïve (FEDN) MDD.This cross-sectional enrolled 1,718 FEDN MDD aged 34.9 ± 12.4 years from First Hospital Shanxi Medical University (Taiyuan, Province, China) September 2016 December 2018. Multivariable binary logistic regression analysis used estimate risk SA. A two-piecewise linear model threshold effects if non-linearity associations existed. Interaction stratified analyses were performed based on sex, education, marital status, comorbid anxiety, psychotic symptoms.Multivariable revealed that positively associated after adjusting for confounders (OR = 1.35, 95% CI: 1.04-1.75, p 0.03). Smoothing plots also showed a nonlinear SA, inflection point being 9.29. On right point, positive detected 3.47, 1.81 6.66, < 0.001), while no significant observed left side 1.14, 0.79 1.66, 0.476).The non-linear exhibited effect MDD. When greater than 9.29, they correlation.
