Abstract The effects of social isolation (SI) during middle age remain unclear, so we tested the hypothesis that SI would lead to an increase in impulsive choice (IC), anxiety‐like behavior, and metabolic dysfunction middle‐aged rats. Male female rats were housed individually or groups four with same‐sex housing mates at 11 months age. Two later, IC behavior was assessed using a delay‐discounting task through novelty‐suppressed feeding (NSF) task. Lastly, glucose tolerance insulin sensitivity following exposure high‐fat diet oral test (OGTT) (ITT). results showed socially isolated displayed more than did group‐housed both sexes. Anxiety disorders are prevalent mental health conditions characterized by significant impairments in daily functioning and social interactions. Despite the effectiveness of pharmacological treatments, challenges such as medication resistance, adverse side effects, high rate relapse necessitate exploration alternative therapies. Recently, electroacupuncture (EA) has garnered attention a promising non-pharmacological intervention for anxiety disorders; however, mechanisms which EA exerts its anxiolytic effects remain poorly understood. This study aims to elucidate role microglial cells anxiety, specifically examining how modulates morphology function within basolateral amygdala (BLA) context induced isolation. Utilizing mouse model isolation-induced we evaluated anxiety-like behaviors through Elevated Plus Maze (EPM) Open Field Test (OFT). Additionally, biochemical analyses immunofluorescence imaging were performed assess expression NADPH oxidase 2 (NOX2), activation markers, levels oxidative stress. Our findings reveal that treatment significantly mitigates mice, correlating with reduction NOX2 BLA microglia decreased reactive oxygen species (ROS). During early life stages, social isolation disrupts the proper brain growth and circuit formation, which is associated with risk of mental disorders cognitive deficits in adulthood. Environmental enrichment (EE) is a significant approach to influencing brain function by altering the environment and changing living conditions has been shown modulate mood-related diseases, including depression. Among elderly, depression particularly prevalent often linked social isolation. However, specific role of EE in isolation-related geriatric remains imprecise. This study was intended explore status exposure uncover its underlying mechanisms. We utilized 19-month-old male C57BL/6J mice, which are equivalent humans aged 50–60 years, induced through After 2 weeks isolation, mice were identified as depressive using sugar preference test then classified into either standard or groups for 4 weeks. Subsequently, conventional indices associated with depression, neurogenesis, neurotrophic factors, neuroinflammation, measured.
