Early-life stress, including maternal separation (MS), increases the vulnerability to develop mood disorders later in life, but underlying mechanisms remain elusive. We report that MS promotes depressive-like symptoms mice at a mature stage of life. Along with this behavioral phenotype, drives reduction GABAB-GIRK signaling and subsequent lateral habenula (LHb) hyperexcitability-an anatomical substrate devoted aversive encoding. Attenuating LHb hyperactivity using chemogenetic tools deep-brain stimulation ameliorates symptoms. This provides insights on strategies alleviate stress-dependent affective behaviors.

Significance Addiction is characterized by compulsive drug use despite negative consequences. In a rat model of methamphetamine self-administration, we found that compulsive-like taking was correlated with alterations in the balance between an increased orbitofrontal cortex-dorsomedial striatal “go” circuit and decreased prelimbic cortex-ventrolateral “stop” only addictive-like subgroup resistant to foot shock punishment paired taking. nonaddictive-like rats, go-stop balance, which similarly changed after repeated self-administration without punishment, normalized toward baseline levels punishment.
