The endocannabinoid system (ECS) is a highly versatile signaling within the nervous system. Despite its widespread localization, functions context of distinct neural processes are very well discernable and specific. This remarkable, question remains as to how such specificity achieved. One key player in ECS cannabinoid type 1 receptor (CB1), G protein–coupled characterized by complexity cell-specific expression, cellular subcellular adaptable regulation intracellular cascades. The tight spatial coupling of synaptic vesicles and voltage-gated Ca2+ channels (CaVs) ensures efficient action potential-triggered neurotransmitter release from presynaptic active zones (AZs). Rab-interacting molecule-binding proteins (RIM-BPs) interact with via RIM other components the machinery. Although human RIM-BPs have been implicated in autism spectrum disorders, little is known about role mammalian transmission. We investigated RIM-BP2-deficient murine hippocampal neurons cultures slices. Short-term facilitation significantly enhanced both model systems. Detailed analysis culture revealed a reduction initial probability, which presumably underlies increased short-term facilitation. Superresolution microscopy an impairment CaV2.1 clustering at AZs, likely alters nanodomains sites thereby affects probability. Endocannabinoids (eCBs) are lipid-based retrograde messengers with a relatively short half-life that produced endogenously and, upon binding to the primary cannabinoid receptors CB1/2, mediate multiple mechanisms of intercellular communication within body. Endocannabinoid signaling is implicated in brain development, memory formation, learning, mood, anxiety, depression, feeding behavior, analgesia, and drug addiction. It now recognized endocannabinoid system mediates not only neuronal communications but also governs crosstalk between neurons, glia, immune cells, thus represents an important player neuroimmune interface. Generation endocannabinoids accompanied by production their congeners, N-acylethanolamines (NAEs), which together N-acylneurotransmitters, lipoamino acids fatty acid amides comprise expanded endocannabinoid/endovanilloid systems. Most these compounds do bind signal via several other pathways involving transient receptor potential cation channel subfamily V member 1 (TRPV1), peroxisome proliferator-activated (PPAR)-α non-cannabinoid G-protein coupled (GPRs) anti-inflammatory, immunomodulatory neuroprotective activities. In vivo generation triggered physiological pathological stimuli specifically brain, fine regulation synaptic strength, neuroprotection, resolution neuroinflammation. Here, we review role intrinsic its therapeutic for treatment neuroinflammation associated synaptopathy.
