The locus coeruleus (LC)-noradrenergic system is the main source of noradrenaline in central nervous and involved intensively modulating pain stress-related disorders (e.g., major depressive disorder anxiety) their comorbidity. However, mechanisms involving LC that underlie these effects have not been fully elucidated, part owing to technical difficulties inherent exploring such a tiny nucleus. novel research tools are now available helped redefine system, moving away from traditional view as homogeneous structure exerts uniform influence on neural activity. Indeed, innovative techniques DREADDs (designer receptors exclusively activated by designer drugs) optogenetics demonstrated functional heterogeneity LC, magnetic resonance imaging applications combined with pupillometry opened way evaluate activity vivo. This review aims bring together data efferent LC-noradrenergic relation its comorbidity anxiodepressive disorders. Acute triggers robust stress response, producing spinal cord-mediated endogenous analgesia while promoting aversion, vigilance, threat detection through ascending efferents. this protective biological fails chronic pain, produces facilitation, anxiety, increased aversive memory, behavioral despair, acting at medulla, prefrontal cortex, amygdala levels. Thus, activation/deactivation specific projections contributes different outcomes shift acute pain.

The locus coeruleus (LC) is a seemingly singular and compact neuromodulatory nucleus that prominent component of disparate theories brain function due to its broad noradrenergic projections throughout the CNS. As diffuse system, noradrenaline affects learning decision making, control sleep wakefulness, sensory salience including pain, physiology correlated forebrain activity (ensembles networks) hemodynamic responses. However, our understanding LC undergoing dramatic shift application state-of-the-art methods reveal many modules provide targeted neuromodulation. Here, we review evidence supporting modular based on multiple levels observation (developmental, genetic, molecular, anatomical, neurophysiological). We suggest concept as and, alongside it, role in diverse must be reconsidered.

This review aims to synthesise a large pre-clinical and clinical literature related hypothesised role of the locus coeruleus norepinephrine system in responses acute chronic threat, as well emergence pathological anxiety. The has widespread projections throughout central nervous system, which act globally modulate arousal states adaptive behavior, crucially positioned play significant modulating both ascending visceral descending cortical neurocognitive information. In response threat or stressor, coeruleus–norepinephrine modulates arousal, alerting orienting functions can have powerful effect on regulation multiple memory systems. Chronic stress leads amplification reactivity subsequent stressors, is coupled with anxiety-like behaviors rodents. While direct vivo evidence for dysfunction humans anxiety remains limited, recent advances high-resolution 7-T magnetic resonance imaging computational modeling approaches are starting provide new insights into characteristics.

To understand what makes sleep vulnerable in disease, it is useful to look at how wake-promoting mechanisms affect healthy sleep. Wake-promoting neuronal activity inhibited during non-rapid-eye-movement (NREMS). However, sensory vigilance persists NREMS animals and humans, suggesting that wake promotion could remain functional. Here, we demonstrate consolidated mouse a brain state with recurrent fluctuations of the neurotransmitter noradrenaline on ∼50-s timescale thalamus. Analysis and interpretation of studies on cognitive affective dysregulation often draw upon the network paradigm, especially Triple Network Model, which consists default mode (DMN), frontoparietal (FPN), salience (SN). DMN activity is primarily dominant during leisure self-monitoring processes. The FPN peaks task involvement exertion. Meanwhile, SN serves as a dynamic “switch” between FPN, in line with demand. In domains, dysfunctions involving are connected to broad spectrum deficits maladaptive behavioral patterns variety clinical disorders, such depression, insomnia, narcissism, PTSD (in case hyperactivity), chronic pain, anxiety, high degrees neuroticism, schizophrenia, epilepsy, autism, neurodegenerative illnesses, bipolar disorder hypoactivity).
