It is becoming increasingly apparent that long-term memory formation relies on a distributed network of brain areas. While the hippocampus has been at center attention for decades, it now clear other regions, in particular medial prefrontal cortex (mPFC), are taking an active part as well. Recent evidence suggests mPFC-traditionally implicated storage memories-is already critical early phases such encoding. In this review, we summarize these findings, relate them to functional importance mPFC connectivity, and discuss role during consolidation with respect different theories storage. Owing its high connectivity areas subserving storage, emerges central hub across lifetime memory, although much still remains be discovered.

Abstract When exploring new environments animals form spatial memories that are updated with experience and retrieved upon re-exposure to the same environment. The hippocampus is thought support these memory processes, but how this achieved by different subnetworks such as CA1 CA3 remains unclear. To understand hippocampal representations emerge evolve during familiarization, we performed 2-photon calcium imaging in mice running virtual compared trial-to-trial dynamics of place cells over days. We find fields rapidly tend shift backwards from remap environment a day later. In contrast, gradually show more stable day-to-day dynamics. These results reflect roles processing familiarization constrain potential mechanisms them.

Tremendous strides have been made in our understanding of the neurobiological substrates memory - so-called "engram". Here, we integrate recent progress engram field to illustrate how neurons transform across "lifespan" a from initial encoding, consolidation and retrieval, ultimately forgetting. To do so, first describe cell-intrinsic properties shape emergence at encoding. Second, highlight these encoding preferentially participate synaptic- systems-level memory. Third, changes during guide neural reactivation facilitate recall. Fourth, mechanisms forgetting, can counteract established consolidation, retrieval. Motivated by experimental results four sections, conclude proposing some conceptual extensions traditional view engram, including broadening cell-type participation within engrams stages. In collection, review synthesizes general principles stages, describes future avenues further understand dynamic engram.

We present here a view of the firing patterns hippocampal cells that is contrary, both functionally and anatomically, to conventional wisdom. argue hippocampus responds efference copies goals encoded elsewhere; it uses these detect resolve conflict or interference between in general. While can involve space, do not encode spatial (or other special types of) memory, as such. also transverse circuits operate an essentially homogeneous way along its length. The apparently different functions parts (e.g. memory retrieval versus anxiety) result from (situational/motivational) inputs on which those perform same fundamental computational operations. On this view, key role iterative adjustment, via Papez-like circuits, synaptic weights cell assemblies elsewhere.

While memories are often thought of as flashbacks to a previous experience, they do not simply conserve veridical representations the past but must continually integrate new information ensure survival in dynamic environments. Therefore, ‘drift’ neural firing patterns, typically construed disruptive ‘instability’ or an undesirable consequence noise, may actually be useful for updating memories. In our view, continual modifications memory reconcile classical theories stable traces with drift. Here we review how updated through recruitment neuronal ensembles on basis excitability and functional connectivity at time learning. Overall, emphasize importance considering static entities, instead flexible network states that reactivate evolve across experience.

Abstract Adult hippocampal neurogenesis (AHN) represents a remarkable form of neuroplasticity that has increasingly been linked to the stress response in recent years. However, hippocampus does not itself support expression different dimensions response. Moreover, main functions are essentially preserved under AHN depletion and adult-born immature neurons (abGNs) have no extrahippocampal projections, which questions mechanisms by abGNs influence supported brain areas far from hippocampus. Significance How do we adaptively switch from perceiving the external world to retrieving goal-relevant internal memories? To tackle this question, used—in a cued-recall paradigm—direct intracranial recordings human hippocampus complemented by high-density scalp electroencephalography (EEG). We found that hippocampal signal ∼500 ms after perceptual cue marks conversion (perceptual) (mnemonic) representations. This sets in motion recall cascade involving posterior parietal and medial prefrontal cortex, revealed via source-localized time-resolved EEG alpha power. Together, these results unveil hippocampal–cortical dynamics supporting rapid flexible memory recall.
