The claustrum (CLA), a subcortical nucleus in mammals, essentially composed of excitatory projection neurons and known for its extensive connections with the neocortex, has recently been associated variety functions ranging from consciousness to impulse control. However, research on CLA challenging due difficulties specifically comprehensively targeting neuronal populations. In various cases, this limitation led inconsistent findings lack reliable data. present work, we describe expression profile Smim32 gene, which is almost exclusively transcribed endopiriform nucleus, as well inhibitory thalamic reticular nucleus. Leveraging unique pattern, developed series Cre- Flippase-expressing knockin BAC transgenic mouse lines different profiles. With these novel tools hand, propose new standards interrogation function.

Behavioral flexibility is the ability to adjust behavioral strategies in response changing environmental contingencies. A major hypothesis field posits that activity of neurons locus coeruleus (LC) plays an important role mediating flexibility. To test this hypothesis, we developed a tactile-based rule-shift detection task which mice responded left and right whisker deflections context-dependent manner exhibited varying degrees switching behavior. Recording spiking from optogenetically tagged LC at millisecond precision during performance revealed prominent graded correlation between baseline flexibility, where higher following rule change was associated with faster new rule. Increasing optogenetic activation accelerated improved performance. Overall, our study provides evidence reveal link

The brain receives constant tactile input, but only a subset guides ongoing behavior. Actions associated with stimuli thus endow them behavioral relevance. It remains unclear how the relevance of affects processing in somatosensory (S1) cortex. We developed cross-modal selection task which head-fixed mice switched between responding to presence visual distractors or using licking movements left right side different blocks trials. S1 spiking encoded stimuli, actions, and direction response not stimuli. Bidirectional optogenetic manipulations showed that sensory-motor activity guided behavior when touch vision was relevant. Our results show its impact on depend actions stimulus.

We have recently identified a group of claustral neurons that continuously maintain information associated with fear-conditioned stimulus (CS) for at least tens seconds, even after the CS has ceased. This “online state” refers to persistent maintenance threat-associated information, enabling it be actively processed threat terminated. state may involve reciprocal interactions brain regions involved in decision-making, motor preparation, and adaptive behavioral responses. If these truly encode online state, their function should remain independent modality or specific defensive behavior exhibited. In this study, we used tone cue monitored freezing trace conditioning, contrast light escape our recent study. During retrieval test subset rostral-to-striatum claustrum (rsCla) exhibited sustained activity response CS, particularly during interval. Importantly, found positive correlation between rsCla magnitude interval, when intervals without were excluded. Thus, appears exhibit characteristics ‘online neurons’ memory following conditioning.

Abstract The claustrum is known for its extensive connectivity with many other forebrain regions, but elongated shape and deep location have made further study difficult. Alzheimer’s disease (AD) is one of the most common neurodegenerative diseases characterized by cognitive deficits and dementia. AD entails predominant pathological characteristics including amyloid beta (Aβ) plaque formation, neurofibrillary entanglements, brain atrophy, which gradually result in dysfunctions. Studies showed that these changes are found a myriad structures, claustrum (CLA), nucleus penetrates deeply into extensively interconnected to various structures. The CLA modulates many aspects functions, with attention, executive function, visuospatial ability, language, memory particular. It also implicated multiple neuropsychiatric disorders, worthy particular attention AD-related impairments. To inspire novel treatment strategies, this review has summarized functionality discriminative dysfunctions AD. And then propose an array potential mechanisms might contribute impairments caused abnormal physiology. Animals are known to exhibit innate and learned forms of defensive behaviors, but it is unclear whether animals can escape through methods other than these forms. In this study, we develop the delayed task, in which male rats temporarily hold information required for future escape, demonstrate that subject extrapolates from past experience without direct its behavioral outcome, does not fall into either two behavior. During holding period, a subset neurons rostral-to-striatum claustrum (rsCla), only when pooled together, sustain enhanced population activity ongoing sensory stimuli. Transient inhibition rsCla during initial part period produces prolonged activity. The transient also attenuates Our data suggest bridges escape-inducing stimuli onset escape.
