Abstract The ventral striatum is a reward center implicated in the pathophysiology of depression. It contains islands Calleja, clusters dopamine D3 receptor-expressing granule cells, predominantly olfactory tubercle (OT). These OT neurons regulate self-grooming, repetitive behavior manifested affective disorders. Here we show that chronic restraint stress (CRS) induces robust depression-like behaviors mice and decreases excitability neurons. Ablation or inhibition these leads to behaviors, whereas their activation ameliorates CRS-induced behaviors. Moreover, has rewarding effect, which diminishes when grooming blocked. Stress coping involves innate and active motivational behaviors that reduce anxiety under stressful situations. However, the neuronal bases directly linking stress, anxiety, motivation are largely unknown. Here, we show acute stressors activate mouse GABAergic neurons in interpeduncular nucleus (IPN). Stress-coping behavior including self-grooming reward sucrose consumption inherently reduced IPN neuron activity. Optogenetic silencing of activation during stress episodes mimicked strategies alleviated anxiety-like behavior. In a model stress-enhanced for seeking, photoinhibition stress-induced sucrose, whereas photoactivation or excitatory inputs from medial habenula potentiated seeking. Single-cell sequencing, fiber photometry, optogenetic experiments revealed stress-activated drive motivated seeking express somatostatin. Together, these data suggest induces motivates to oppose as an anxiolytic stress-coping mechanism.

Re-examining the relationship between neuropeptide systems and neural circuits will help us to understand more intensively critical role of neuropeptides in brain function as responsible for specific functions are gradually revealed. Gastrin-releasing peptide receptors (GRPRs) Gαq-coupling widely distributed brain, including hippocampus, amygdala, hypothalamus, nucleus tractus solitarius (NTS), suprachiasmatic (SCN), paraventricular hypothalamus (PVN), preoptic area (POA), preBötzinger complex (preBötC), etc., implying GRP/GRPR system is involved modulating multiple functions.
