Our objective was to explore the patterns of resting-state network (RSN) connectivity alterations and investigate how influences individual-level connections on cognition varied across clinical stages without assuming a constant relationship. 108 PD patients with continuum cognitive decline (PD-NC = 46, PD-MCI 43, PDD 19) 34 healthy controls (HCs) underwent functional MRI neuropsychological tests. Independent component analysis (ICA) graph theory analyses (GTA) were employed RSN connection changes. Additionally, stage-dependent differential impact communication performance examined using sparse varying coefficient modeling. Compared HCs, dorsal attention (DAN) sensorimotor (dSMN) central networks decreased in PD-NC stage, while lateral visual (LVN) emerged as dementia. cerebellum (CBN) increased stages. Cognitive impairment (CI) is common in α-synucleinopathies, i.e., Parkinson's disease, Lewy bodies dementia, and multiple system atrophy. We summarize data from systematic reviews/meta-analyses on neuroimaging, neurophysiology, biofluid genetic diagnostic/prognostic biomarkers of CI α-synucleinopathies. Diagnostic include atrophy/functional neuroimaging brain changes, abnormal cortical amyloid tau deposition, cerebrospinal fluid (CSF) Alzheimer's disease (AD) biomarkers, rhythm slowing, reduced cholinergic glutamatergic increased GABAergic activity, delayed P300 latency, plasma homocysteine cystatin C decreased vitamin B12 folate, CSF/serum albumin quotient, serum neurofilament light chain. Prognostic regional atrophy, CSF Val66Met polymorphism, apolipoprotein-E ε2 ε4 alleles. Some AD/amyloid/tau may diagnose/predict but single, validated lack. Traumatic brain injuries (TBIs) pose a significant health concern among the elderly population, influenced by age-related physiological changes and prevalence of neurodegenerative diseases. Understanding biomechanical dimensions TBIs in this demographic is vital for developing effective preventive strategies optimizing clinical management. This comprehensive review explores intricate biomechanics elderly, integrating medical aging studies, experimental head tissues, numerical simulations. Research reveals that global atrophy normal occurs at annual rates -0.2% to -0.5%. In contrast, diseases such as Alzheimer's, Parkinson's, multiple sclerosis are associated with significantly higher atrophy.
