Background The retrograde endocannabinoid (eCB) pathway is closely associated with the etiology of major depressive disorder (MDD) at both pathophysiological and genetic levels. This study aimed to investigate potential role mutations in eCB underlying mechanisms Han Chinese patients MDD. Methods A total 96 drug-naïve first-episode MDD 62 healthy controls (HCs) were recruited. Whole-exome sequencing was performed identify gene mutation profiles Results filtered focus on low-frequency variants rare (minor allele frequencies <0.05) related phenotypes. Enrichment analyses for 146 selected genes examine pathways which most significant enrichment occurred. protein–protein interaction (PPI) network analysis explore biological functions pathway. Finally, based current literature, a preliminary conducted effect function this Our identified (15.02%) depression-related when compared HCs, 37 enriched signaling Seven hub mitochondrial function, including Complex I (NDUFS4, NDUFV2, NDUFA2, NDUFA12, NDUFB11) protein (PARK7) enzyme (DLD) regulation oxidative stress. Conclusion These results indicate that represent etiological factors pathogenesis

The endocannabinoid system is found throughout the central nervous system, and its cannabinoids receptor 1 critical in preventing neurotoxicity caused by N-methyl-D-aspartate activation (NMDARs). activity of NMDARs places demands on endogenous to regulate their calcium currents. Endocannabinoids keep NMDAR within safe limits, protecting neural cells from excitotoxicity. Cannabinoids are remembered deliver this outcome repressing presynaptic glutamate discharge or obstructing postsynaptic NMDAR-managed flagging pathways. must exert a negative influence proportional strength signaling for such control be effective. goal paper draw attention towards neuroprotective mechanism constituents Cannabis sativa against NMDA-induced excitotoxic result. Phytochemical investigation cannabis flowers led isolation nine secondary metabolites. A spiro-compound, Cannabispirenone A, which treatment prior NMDA exposure significantly increases cell survival while decreasing ROS production, lipid peroxidation, intracellular calcium. Our findings showed that compound neuroprotection insult, has antioxidative properties, increased cannabinoid expression, may involved pathway neuroprotection.

The endocannabinoid system (ECS) is a critical important neuromodulatory that interacts with many neurohormonal and neurotransmitter systems in the brain. It plays pivotal role emotional responses mood regulation. ECS related psychotic disorders, depression, anxiety autism. In this study, we aimed to investigate whether there any relationship between N-acylethanolamine levels bipolar disorder.Seventy-nine patients disorder diagnosis, who are euthymic period, were included study. Clinical characteristics, symptoms serum compared. Endocannabinoid evaluated using liquid chromatography-tandem mass spectrometry. In total of 79 patients, 44 (55.69%) females 35 (44.30%) males. mean age was 42.40 ± 1.10 years. Palmitoylethanolamide (PEA) higher oleoylethanolamide 2-arachidonyl glycerol lower had at least one depressive episode during their life-time illness than no while arachidonyl ethanolamide unchanged.PEA correlated history frequency episodes disorder.

The recent discovery of N-acyl taurines (NATs) as a class endogenous bioactive lipids and the perspective their possible pharmacological applications stimulated development mass spectrometry-based methods for quantitative measurements in biological tissues fluids. We report here first time procedure validated both liver surrogate matrix neat solvent (MeOH) based on UPLC-ESI-QqQ analysis identification quantification NATs tissue extracts. LC-MS method was five representative lipid analogues, including saturated, monounsaturated polyunsaturated species, namely N-palmitoyl taurine (C16:0 NAT), N-oleoyl (C18:1 N-arachidonoyl (C20:4 N-docosanoyl (C22:0 NAT) N-nervonoyl (C24:1 evaluated specificity, linearity, effect, recovery, repeatability intermediate precision accuracy. MeOH by internal standard approach (d4-C20:4 showed excellent linearity range 1-300 ng/ml with R always ≥ 0.9996 all NATs; intra-day inter-day accuracy were within acceptable range. These findings prompt further studies disclose function various members this different peripheral tissues.
