Abstract Rising antibiotic resistance among periodontal pathogens poses a major challenge to treatment outcomes. This work investigates the possible combinatorial potential of AgNPs synthesized using glycyrrhizin, natural biosurfactant, combat biofilm‐forming bacteria isolated from periodontitis patients. In silico findings revealed that glycyrrhizin had higher binding affinity score ‐9.102 compared doxycycline (−3.599) for gtfB , plays an indispensable function in biofilm integrity. were and optimized utilizing design experiments, considering factors such as pH, temperature, incubation time concentration variables. while The resulting GL‐AgNPs characterized by different analytical techniques FTIR, XRD, DLS, SEM. nanoparticles non‐spherical, uniform average size 72 nm, zeta potentials ranged −28 mV − 42 mV. Multidrug resistance in cancer stem cells (CSCs) is a major barrier to chemotherapy; hence, developing CSC-specific targeted nanocarriers for efficient drug delivery critical. In this study, monodisperse hollow-structured MnO2 (H-MnO2) with mesoporous shell was created delivery. An effective therapeutic compound isoliquiritigenin (ISL) confirmed inhibit the lung stem-cell phenotype by natural screening based on integrated microfluidic devices. The resultant H-MnO2 showed high drug-loading content of potent CSC-targeting ISL and near-infrared fluorescent dye indocyanine green (ICG). addition, successively modified hyaluronic acid (HA) enhance targeting CSCs CD44 expression levels. H-MnO2@(ICG + ISL)@HA nanocomposites displayed promising chemotherapeutic photothermal treatment capabilities, as well NIR-triggered release, which excellent CSC-killing effects tumor inhibition efficacy. Meanwhile, development effectively restrained phototherapy prominent chemotherapy without obvious side after tail vein injection vivo. summary, prepared accomplished synergistic therapy that targets CSCs, offering versatile platform diagnosis treatment.

Chronic inflammation is associated with a prolonged increase in various inflammatory factors. According to clinical data, it can be linked both cancer and autoimmune diseases the same patients. This raises critical question of how chronic relates seemingly opposing - tumors, which there immunosuppression, diseases, over-activation immune system. In this review, we consider as prerequisite for suppression an increased likelihood damage. We also discuss potential disease-modifying therapies targeting inflammation, helpful autoimmunity. As one of the most common malignancies, colorectal cancer (CRC) usually starts with a benign lesion and accumulates DNA damage as it progresses to full-fledged cancer. Glycyrrhizin (GL) has been found alleviate tumor growth inflammation, while role GL influences response (DDR) in remains unclear. exposure significantly reduced cell colony formation viability concomitant increase fragmentation CRC, meanwhile induced apoptosis by activating caspase-3. Moreover, cycle arrest CRC cells at S phase, which was associated decreased cyclin D1 vitro. treatment ameliorated promoted DDR vivo. AtGUS, identified in the genome of Aspergillus terreus Li-20, can hydrolyze glycyrrhetinic (GL) into acid monoglucuronide (GAMG) and (GA). However, poor substrate specificity AtGUS often limits its further application. In this study, highly conserved Glu416 Glu507 residues were as catalytic sites while Glu158, Asp163, Arg565, which form hydrogen bonds with substrate, play critical roles enzymatic activity, Arg565 being particularly important. When on loop 8 surface TIM barrel was mutated to glutamate, GAMG became sole product, yield from GL conversion up 85%. hydrolysis reaction terminated, 7.32-fold higher than that wild enzyme. Molecular docking dynamic simulation analyses revealed affinity for mutant R565E (Km = 0.192 mmol/L) improved, but reduced efficiency toward (kcat 5.515 ± 0.07 mmol–1 L s–1). The objective of the present study was to elucidate mechanism by which glycyrrhizin enhances antitumor activity cisplatin in non-small cell lung cancer. Initially, A549 cells were treated with different concentrations (0.25-8 mM) or (10-160 µM) for 48 h investigate effect combined on vitro. Subsequently, divided into control (untreated), CP (20 µM cisplatin), GL (2 mM glycyrrhizin) and + 2 groups underlying glycyrrhizin. After incubation, viability colony-forming ability assessed using MTT colony formation assays. Apoptosis levels cycle progression analyzed flow cytometry western blotting used evaluate apoptosis- cycle-related proteins. Additionally, comet assays DNA damage relevant results demonstrated both individually reduced a concentration-dependent manner. In addition, analysis that, comparison alone, markedly increased protein expression B-cell lymphoma 2-associated X protein, cleaved-caspase-3/caspase-3, γH2AX, phosphorylated-checkpoint kinase 1 phosphorylated-p53/p53, while notably reducing 2, cyclin D1, cyclin-dependent 4. findings indicate that cancer modulating apoptosis.
