Slow transit constipation (STC) is a prevalent gastrointestinal condition with slow transit, and some probiotics can effectively relieve constipation, but the exact mechanisms have not been fully understood. In this study, we evaluate impact of Lactiplantibacillus plantarum GUANKE (GUANKE) on diphenoxylate-induced speculate underlying in mouse model. Administration L. alleviated indexes, including defecation time, fecal output water content, ratio. addition, restored protein expression constipation-related intestinal factors (aquaporins (AQPs) interstitial Cajal cells (ICCs)) colon tissues measured using immunofluorescence staining; regulated neurotransmitters hormones, such as increased levels 5-hydroxytryptamine, substance P, motilin; decreased vasoactive peptide nitric oxide serum, by an ELISA. 16S rRNA correlation analysis feces indicated that administration reduced constipation-induced Prevotella enrichment suggested potential contribution to STC mice. had no effect short-chain fatty acids (SCFAs) cecum content. This study revealed may alleviate mice through regulating neurotransmitter hormone release altering specific bacterial taxa, rather than affecting SCFAs diversity microbiota gut. Further research needed confirm if findings observed will be consistent other animal studies or clinical trials.

Background: Amyotrophic lateral sclerosis (ALS) is a rapidly progressing and rare neurodegenerative disease. Therefore, evaluating the risk factors affecting survival of patients with ALS crucial. Constipation, common but overlooked symptom ALS, can be effectively managed. It currently unknown whether constipation contributes to progression ALS. Objectives: This study aimed investigate association between development using novel overlap-weighted (OW) method enhance robustness reliability results. Design: prospective matching nested case-control (NCC) was conducted within an ongoing cohort at Second Hospital Hebei Medical University. Baseline data were collected from meeting inclusion exclusion criteria, as exposure factor. A 9-month follow-up conducted, death endpoint event. Methods: We primarily used OW in NCC studies examine survival. Weighted Cox proportional hazards model assess associated overall Survival differences two groups analyzed Kaplan-Meier’s plots log-rank tests. Finally, bioinformatic analysis explored pathways constipation. Results: Among 190 included, prevalence 50%. Patients exhibited faster disease ( p < 0.001), positive correlation severity rate r = 0.356, 0.001). The group had poorer before after (log-rank test, 0.0001). In 114 patients, factor for both (hazard ratio (HR) 5.840, 95% confidence interval (CI) 1.504–22.675, 0.011) (HR 5.271, CI 1.241–22.379, 0.024) OW. Conclusion: Constipation individuals reduced rates, potentially through peroxisome proliferator-activated receptor pathway.

This study aimed to understand the genetic and metabolic traits of a Lactiplantibacillus plantarum JS21 strain its probiotic abilities through laboratory tests computer analysis. L. was isolated from traditional fermented food known as “Jiangshui” in Hanzhong city. In this research, complete makeup determined using Illumina PacBio technologies. The genome consisted 3.423 Mb circular chromosome five plasmids. It found contain 3023 protein-coding genes, 16 tRNA 64 rRNA operons, 40 non-coding RNA 264 pseudogenes, six CRISPR array regions. GC content 44.53%. Additionally, harbored three prophages. evolutionary relationship collinearity were compared with other strains. resistance genes identified inherent. Enzyme involved Embden–Meyerhof–Parnas (EMP) phosphoketolase (PK) pathways detected, indicating potential for facultative heterofermentative pathways. possessed bacteriocins plnE/plnF polyketide terpenoid assembly, possibly contributing antibacterial properties against Escherichia coli (ATCC 25922), (K88), Staphylococcus aureus (CMCC 26003), Listeria monocytogenes (CICC 21635). Furthermore, carried Na+/H+ antiporters, F0F1 ATPase, stress which may account ability withstand simulated conditions human gastrointestinal tract vitro. high hydrophobicity cell surface suggested intestinal colonization. Overall, exhibited evidenced by experiments computational analysis, suggesting suitability dietary supplement.

Citrus is an important genus in the Rutaceae family, and citrus peels can be used both food herbal medicine. However, bulk of are discarded as waste by fruit processing industry, causing environmental pollution. This study aimed to provide guidelines for rational effective use elucidating volatile nonvolatile metabolites within them using metabolomics based on headspace-gas chromatography–ion mobility spectrometry ultra-high-performance liquid chromatography–Q-Orbitrap high-resolution mass spectrometry. In addition, antioxidant activities were evaluated DPPH radical scavenging, ABTS ferric reducing power. total, 103 53 identified characterized. Alcohols, aldehydes, terpenes constituted 87.36% metabolites, while flavonoids carboxylic acids accounted 85.46% metabolites. Furthermore, (Z)-2-penten-1-ol, L-pipecolinic acid, limonin characteristic components reticulata Blanco cv. Ponkan (PK), C. 'Unshiu' (CLU), 'Wo Gan' (WG), respectively. Principal component analysis partial least squares discriminant indicated that 'Chun Jian' (CJ), PK, CLU, 'Dahongpao' (DHP) clustered together. DHP a traditional Chinese medicine documented Pharmacopoeia, suggesting chemical compositions CJ, CLU may also have medicinal values similar those DHP. Moreover, DHP, 'Ai Yuan 38'(AY38), 'Gan Ping'(GP), 'Qing Jian'(QJ) displayed better activities, recommending their additives cosmetics food. Correlation suggested some polyphenols including tangeritin, nobiletin, skullcapflavone II, genistein, caffeic isokaempferide potential compounds peel. The results this deepen our understanding differences different peel varieties ultimately guidance full peels.
