Abstract Nonalcoholic fatty liver disease (NAFLD) encompasses a broad range of conditions, commencing with simple steatosis and progressing to non‐alcoholic steatohepatitis, the possibility further deterioration into fibrosis, cirrhosis, ultimately, hepatocellular carcinoma. Unfortunately, there is currently no approved medication for treating NAFLD‐associated steatosis. This underscores need improved therapeutic approaches that can modulate lipid metabolism halt transition from chronic disease. Our previous studies have demonstrated apoptotic vesicles (apoVs), which are produced during apoptosis, show great potential in regulating homeostasis. However, whether they ameliorate NAFLD unknown. In our research, apoVs derived platelets (PLT‐apoVs) as well mesenchymal stem cells (MSC‐apoVs) were used treat NAFLD. The results showed PLT‐apoVs exhibited superior effects diminishing accumulation induced by high‐fat diet than MSC‐apoVs. Through proteomic analysis, we defined validated apolipoprotein A‐II (APOA2) regulator apoV‐mediated MSC adipogenesis, could be target enhance apoV biomedical field. Introduction The Constitutive Androstane Receptor (CAR) (NR1I3), a pivotal member of the xenosensor family, plays key role in hepatic detoxification xenobiotic and endobiotic chemicals through induction expression drug-metabolizing enzymes transporters. CAR’s involvement extends beyond detoxification, influencing gluconeogenesis, lipogenesis, bile acid regulation, cellular processes such as proliferation, tissue regeneration, carcinogenesis. This review explores CAR regulation by various factors, highlighting its mediating metabolic changes induced environmental contaminants. Methods A literature search was conducted to identify all articles on PubMed website which CAR-contaminant CAR-hepatic steatosis relationship is analyzed both vitro vivo models. Results Numerous contaminants, perfluorooctanoic (PFOA), Zearalenone mycotoxin, PCB, triazole fungicide propiconazole can activate nuclear receptors contributing development increased de novo decreased fatty oxidation, lipid uptake, gluconeogenesis. Bisphenol analogs and their derivatives have been identified in human tissue our living environment. There are major concerns over exposure to bisphenol analogs, especially the low-dose- mixture-related toxicities, as they considered potential endocrine-disrupting chemicals that may cause adverse effects multiple organ systems. The liver is a critical responsible for an array of functions, e.g., metabolism, immunity, digestion, detoxification vitamin storage, addition being leading chemical target site. In this literature review species, we discussed metabolism liver, which was followed by discussions analog-induced toxicity various including humans, rodents (mice rats) other species (chicken, pig, sheep, etc.). Abstract Obesity, diabetes, and their cardiovascular hepatic comorbidities are alarming public health issues of the twenty-first century, which share mitochondrial dysfunction, oxidative stress, chronic inflammation as common pathophysiological mechanisms. An increasing body evidence links combined exposure to multiple environmental toxicants with occurrence severity metabolic diseases. Endocrine disruptors (EDs) ubiquitous chemicals or mixtures persistent deleterious effects on living organisms beyond endocrine system impairment; in particular, those known metabolism-disrupting (MDCs), increase risk pathologies adult organism its progeny. Being largely lipophilic, MDCs mainly target adipose tissue elicit dysfunction by interfering bioenergetics, biogenesis, dynamics and/or other functions. Plastics, when broken down into micro- nano-plastics (MNPs), have been detected several human tissues, including liver. The harmful interplay between inflammatory redox processes, mutually interact a positive feed-back loop, hence term ("OxInflammation"), occurs both at systemic organ level. In liver tissue, oxinflammation contributes progression dysfunction-associated steatotic disease (MASLD). Moreover, it has reported that individuals MASLD may be more susceptible (mainly, related mitochondria) EDs/MDCs MNPs play role development disease.
