Wuzhuyu decoction (WZYD) is a well-known classic traditional Chinese medicine prescription and has been widely used to treat headache, nausea, vomiting, insomnia, etc. However, little published information available about its safety. Our aim was investigate the acute subacute oral toxicity of WZYD extract in rats following technical guidelines from China's National Medical Products Administration (NMPA) for single repeated doses studies drugs. Acute assessed via administration at 4 g/kg three times within day followed by 14-day observation period. To evaluate toxicity, were orally administered with 0, 0.44, 1.33, 28 days. The items examined included clinical signs, body weight, food consumption, hematological biochemical parameters, bone marrow smear, organ index, histopathology. After 12 (3 × g/kg) extract, no mortality toxic effects observed during In study, did not cause any significant treatment-related abnormality each item rats, so adverse effect level (NOAEL) days considered be g/kg, which approximately 80-fold proposed dosage.

Migraines constitute a common neurological and headache disorder affecting around 15% of the world’s population. In addition to other mechanisms, neurogenic neuroinflammation has been proposed play part in migraine chronification, which includes peripheral central sensitization. There is therefore considerable evidence suggesting that inflammation intracranial meninges could be key element calcitonin gene-related peptide (CGRP), leading sensitization trigeminal meningeal nociceptors migraines. are several studies have utilized this approach, with strong focus on using inflammatory animal models. Data from these show process involves trigeminovascular afferent nerve terminals. Further, by applying wide range different pharmacological interventions, insight gained pathways involved. Importantly, we discuss how models should used care it important evaluate outcomes light pathology.
