Major depressive disorder (MDD) is one of the most common mental health conditions, characterized by pervasive and persistent low mood, self-esteem, a loss interest or pleasure in activities that are typically enjoyable. Despite decades research into etiology pathophysiological mechanisms depression, therapeutic outcomes for many individuals remain less than expected. A promising new area focuses on stress-induced neuroinflammatory processes, such as excessive activation crosstalk microglia astrocytes central nervous system under stress, well elevated levels pro-inflammatory cytokines, which closely linked to onset progression depression. This review summarizes through neuroinflammation induces promotes development also highlights effective roles small molecules with anti-inflammatory activity treatment MDD. Understanding specific further impacts using technologies single-cell RNA sequencing elucidate subtypes interactions great importance developing more strategies

Antioxidant plasticity-promoting molecules, such those Nrf2–HO-1, BDNF–TrkB, cyclic AMP–CREB pathways, could protect depression, while glycogen synthase kinase-3β nuclear factor κB oppose these actions, thus depressive-like behaviours. Since also endowed tumorigenic atherogenic potential, balance between benefits harms must be taken into account designing novel drugs aiming at intracellular Nrf2.

Abstract Aim We investigated the effects and target of gastrodin (GAS) for treating depression through network pharmacology combined with experimentation. Methods The therapeutic signal GAS were analyzed by pharmacology. Depression in mice was mimicked a chronic unpredictable mouse stress (CUMS) model. Through open field, elevated plus maze, forced swimming, tail suspension tests, on CUMS behaviors examined, levels neurotransmitters detected. histopathological changes assayed H&E IHC staining, protein expressions detected Western blotting. Small molecule‐protein docking molecular dynamics experiments conducted to simulate binding mode between Caspase‐3. Results Network pharmacological analysis revealed that Caspase‐3 action GAS. could improve depression‐like mice, elevate their neurotransmitter levels, ameliorate nerve cell injury, inhibit expression. After knocking out Caspase‐3, inhibited. Molecular simulation small found bound at SER25, inhibiting maturation activation Conclusion find can act as inhibitor, which improves injury Caspase‐3‐mediated apoptosis.

Depression is a psychiatric disorder and the treatment of depression an urgent problem that need to be solved. Gastrodin (GAS) Traditional Chinese Medicine from orchid used for neurological diseases, including depressive disorders. To assess effect GAS on gut microbiota mice, we established chronic unpredictable mild stress (CUMS)-induced mouse model, was administered one group mice. Animal behavior experiments were detect depressive-like behaviors, 16 S rRNA gene analysis applied each group. All raw sequences deposited in NCBI Sequence Read Archive under accession number SRP491061. significantly improved behaviors as well diversity abundance microbiota. The CUMS-GAS different degrees compared with CUMS linear discriminant (LDA) showed makeup mice changed dramatically group, Bacteroides (LDA = 3.94, P < 0.05) enriched at genus level. In comparison had greater concentration numbers Lactobacillus, Corynebacterium, Staphylococcus, Bacteroides, Psychrobacter, Alistipes. Our results suggested impacted microbial composition gut. research indicated dysbiosis may affected by treatment, which CUMS-induced model depression.

Inflammation-induced oligodendrocyte death and CNS demyelination are key features of multiple sclerosis (MS). Inflammation-triggered endoplasmic reticulum (ER) stress oxidative promote tissue damage in MS its preclinical animal model, experimental autoimmune encephalitis (EAE). Compound AA147 is a potent activator the ATF6 signaling arm unfolded protein response (UPR) that can also induce antioxidant through activation NRF2 pathway neuronal cells. Previous work showed protects tissues against ischemia/reperfusion and/or activation; however, therapeutic potential neuroinflammatory disorders remains unexplored. Here, we demonstrate ameliorated clinical symptoms EAE reduced ER stress, loss, demyelination. Additionally, suppressed T cells without altering peripheral immune response. Importantly, significantly increased expressions Grp78, an target gene, oligodendrocytes, while enhancing levels Grp78 as well Ho-1, microglia. In cultured promoted nuclear translocation ATF6, but not NRF2. Intriguingly, altered microglia profile, possibly by triggering pathway. was therapeutically beneficial during acute stage mice lacking indicating protection primarily involves these Overall, our results suggest opportunity for promoting survival regulating status distinct mechanisms.
