Objective: This study aimed to investigate methodologies for the extraction and purification of polysaccharides from Rosa roxburghii Tratt fruits their impact on various cellular processes in prostate cancer DU145 cells, including survival rate, migration, invasion, cell cycle, apoptosis. Results: Compared control group, polysaccharide exhibited a significant reduction viability, invasion rates cells time- dose-dependent manner within polysaccharide-treated groups. Additionally, it effectively arrested cycle at G0/G1 phase by downregulating expressions CDK-4, CDK-6, Cyclin D1. Furthermore, induced apoptosis upregulating Caspase 3, 8, 9, BAX. Methods: Polysaccharides were extracted sourced Yunnan, China. Extraction decolorization methods optimized using response surface methodology, based single-factor experiment. Polysaccharide was carried out DEAE-52 cellulose Sephadex G-100 column chromatography. The optimal dosage R. affecting determined CCK-8 assay. Cell migration assessed transwell scratch assays. Conclusions: polysaccharides, consisting D-mannose, L-rhamnose, N-acetyl-D-glucosamine, D-galacturonic acid, D-glucose, D-galactcose, D-xylose, L-arabinose, L-fucose, possess ability hinder proliferation, while inducing through modulation relevant gene expressions.
