Metabolic cancers are defined by metabolic reprogramming. Although this reprograming drives rapid tumour growth and invasion, it also reveals specific vulnerabilities that can be therapeutically exploited in cancer therapy. A novel form of programmed cell death, known as disulfidptosis, was identified last year; cells with high SLC7A11 expression undergo disulfidptosis when deprived glucose. Natural products have attracted increasing attention shown potential to treat through diverse mechanisms.

Abstract Owing to the developments in various therapeutic modalities, cancer treatment has come a long way, including discovery of anticancer drugs, innovations delivery technology, and increased personalization treatments. Despite this, head neck (HNC) is that eluded current conventional To improve quality life preserve vital organ function patients with HNC, there need for research into regimes would reduce toxicity modalities. The use single approach not been enough completely eradicate this malignancy. Therefore, adjuvants combinatorial approaches, using molecules from natural compounds have no or minimal side effects, growing area research. One objective review was clarify potentiality novel strategies such as phytochemicals chemotherapy radiotherapy, nano-formulation delivery. Another delineate associated challenges clinical application these therapies HNC. Possible overcoming critical issues HNC are also discussed.

Background/Objectives: Head and neck cancer is one of the most common cancers globally, with high mortality significant treatment-related side effects. Conventional therapies, including surgery, radiotherapy, chemotherapy, have improved survival but often serious consequences for patients’ quality life. For this reason, there growing interest in complementary therapies such as acupuncture, herbal medicine, cannabinoids, traditional Chinese mind-body therapies. Methods: This review was conducted through a systematic analysis scientific literature available on PubMed Scopus, selecting studies about use alternative patients head according to strict criteria. Results: Acupuncture has shown benefits management xerostomia dysphagia, while some medicines potential anticancer effects, although limitations related bioavailability. Vitamins antioxidants showed mixed results: suggest protective effect, others report possible increased risk progression. Cannabinoids are controversial topic, palliative also higher cancer. Traditional medicine yoga, positive effects well-being, their direct impact progression remains uncertain. Conclusions: Alternative could be useful support managing symptoms improving life However, solid evidence effectiveness safety still lacking. Rigorous clinical needed assess therapeutic define safe integration into multidisciplinary management.

Oral Lichen Planus (OLP) is a chronic inflammatory disorder that may progress to Squamous Cell Carcinoma (OSCC). Lipid metabolism dysregulation has been implicated in tumor development and immune response modulation. This study aims explore the role of lipid metabolism, particularly lipids diacylglycerol (DAG), triacylglycerol (TAG), phosphatidylcholine (PC), progression from OLP OSCC, identify potential therapeutic targets for prevention treatment. We performed Mendelian randomization (MR) analysis investigate causal relationships between risk OSCC. Differential gene expression was conducted key genes related metabolism. The interactions species were examined using drug databases (DrugBank, DGIdb, TCMSP) candidates. Enrichment signaling pathways, including PPAR signaling, also understand underlying mechanisms. Our MR revealed DAG exerts protective effect (OR < 1), but its shifts factor OSCC > potentially by altering microenvironment. TAG PI plays critical tumorigenesis. Gene identified several metabolism-related genes, SLC27A6, FABP3, FABP4, ADIPOQ, PLIN1, whose differed highlighting their importance progression. These enriched pathway, suggesting involvement growth Potential candidates, such as palm acid (PA), Imatinib, Curcumin, through drug-repurposing strategies. crucial Targeting pathways DAG, TAG, PI, offer promising strategies early diagnosis intervention. provides novel insights into molecular mechanisms OLP-to-OSCC suggests natural compounds, future clinical applications. Further research needed validate these findings settings. Not applicable.

Oxidative stress serves as both a driver and result of redox metabolism across diverse physiological pathological states, including cancer. Head neck squamous cell carcinoma (HNSCC), the sixth most prevalent malignancy worldwide, is no exception. HNSCC strongly linked to modifiable external risk factors such tobacco smoking, alcohol consumption, high-risk human papilloma (HR-HPV) infection. These are associated with elevated oxidative stress, which contributes carcinogenesis through DNA damage, chronic inflammation, dysregulation signaling pathways. Current treatment options for have limitations burden side effects. Studies been conducted on potent dietary antioxidants prevention adjunctive HNSCC.
