Abstract Alzheimer’s disease is a neurodegenerative disorder and characterized by amyloid beta accumulation, synaptic dysfunction, oxidative stress, lacks effective therapies. Caloric restriction mimetics such as fisetin chlorogenic acid, natural polyphenols with antioxidant autophagy-inducing properties, show promise in mitigating age-related diseases. This study investigates their neuroprotective effects against induced toxicity differentiated human neuroblastoma SHSY5Y cells. Amyloid exposure disrupted redox homeostasis, impaired autophagy, mitochondrial exacerbating neuronal degeneration. Fisetin acid treatments reversed these deleterious restoring balance, suppressing reactive oxygen species upregulating critical enzymes like SOD1, GSR, catalase. These compounds also attenuated mitophagy via reduced PINK1 expression restored fusion Mfn2. Autophagy-related pathways were significantly modulated, evidenced increased AMPK decreased mTOR mRNA levels, alongside elevated of ATG101, ATG13, ULK1, P62 ATG5 levels. Docking studies revealed binding CGA within the pockets FKBP12 supporting interaction. Furthermore, improved integrity PSD95 synaptophysin reducing acetylcholinesterase expression. findings highlight potential ameliorating through autophagy activation, preservation, function enhancement. Chlorogenic acid (CGA) is a type of polyphenol compound found in rich concentrations many plants such as green coffee beans. As an active natural substance, CGA exerts diverse therapeutic effects response to variety pathological challenges, particularly conditions associated with chronic metabolic diseases and age-related disorders. It shows multidimensional functions, including neuroprotection for neurodegenerative disorders diabetic peripheral neuropathy, anti-inflammation, anti-oxidation, anti-pathogens, mitigation cardiovascular disorders, skin diseases, diabetes mellitus, liver kidney injuries, anti-tumor activities. Mechanistically, its integrative functions act through the modulation anti-inflammation/oxidation homeostasis. can thwart inflammatory constituents at multiple levels curtailing NF-kB pathways neutralize primitive factors, hindering propagation, alleviating inflammation-related tissue injury. concurrently raises pivotal antioxidants by activating Nrf2 pathway, thus scavenging excessive cellular free radicals. elevates AMPK maintenance restoration homeostasis glucose lipids. Additionally, neuromodulation targeting neuroreceptors ion channels. In this review, we systematically recapitulate CGA’s pharmacological activities, medicinal properties, mechanistic actions potential agent. Further studies defining specific molecules, improving bioavailability, validating clinical efficacy are required corroborate CGA.

ABSTRACT Phenolic compounds, abundantly found in medicinal herbs, may promote health by modulating the gut microbiota. Yarrow is a traditional remedy for digestive and non‐gastrointestinal diseases with unexplored interaction microbes. Prebiotic demand is increasing due to growing concerns about human gut health and well-being. Recently, agri-food by-products have been explored as a potential source of prebiotic ingredients. This study analysed the chemical composition freeze-dried coffee silverskin extract (FSE) evaluated its impact on Lacticaseibacillus paracasei subsp. BAA-52 ATCC growth. The FSE was obtained by multi-frequency multimode modulation ultrasonic vibration. Soluble dietary fiber accounted for 15.2 % extract, being mainly composed uronic acids (58 mol%) glucose (13.4 mol%). HPLC-DAD-MS-ESI+ HPLD-DAD analyses revealed phenolic-rich (mainly caffeoylquinic acids), well presence caffeine, 5-hydroxymethylfurfural. probiotic incubated in De Man, Rogosa, Sharpe broth supplemented with different concentrations (1–4 %, w/v). 2 4 (w/v) significantly enhanced L. subs. growth decreased pH medium, indicating ingredient.
