UCP2 is an uncoupling protein homolog to UCP1. Unlike UCP1, which participates in non-shivering thermogenesis by oxidative phosphorylation (OXPHOS), does not perform a canonical H+ leak, consuming the protonmotive force (Δp) through inner mitochondrial membrane. The biological role elusive. It can counteract stress, acting with “mild uncoupling” process reduce ROS production, and, fact, activities are related inflammatory processes, triggering pathological conditions. However, Δp dissipation activity reduces ATP production and rewires bioenergetic metabolism of cells. In all likelihood, works as carrier metabolites four carbon atoms (C4), reversing anaerobic glycolysis-dependent catabolism OXPHOS. Indeed, catalysis dual mode: mild OXPHOS metabolite C4 exchange mitochondria. vivo, features biology mitochondria promote healthy ageing, increased lifespan, assure cerebro- cardiovascular protection. conditions responsible for insulin secretion suppression dependent on activity. Abstract Diabetic retinopathy (DR) represents a significant and serious complication associated with diabetes mellitus (DM), often resulting in considerable visual impairment or even blindness. The intricate pathological processes underlying DR complicate the effectiveness of current treatment modalities. Studies have highlighted potential natural products via several beneficial effects including anti-inflammatory, antioxidant, anti-neovascular, anti-apoptotic properties. Flavonoids, saponins, saccharides, alkaloids exhibited various vivo vitro studies. However, clinical utilization these compounds is hindered by issues such as inadequate specificity, low bioavailability, toxicity. Therefore, there pressing need for rigorous studies to confirm efficacy preventing mitigating progression DR.

Introduction Periodontitis is a chronic inflammatory disease closely associated with mitochondrial dysfunction. Uncoupling protein 2 (UCP2), located in the inner membrane of mitochondria, reduces potential (MMP) and adenosine triphosphate (ATP) synthesis by promoting proton leakage across membrane. This leads to decreased energy metabolism efficiency, impairing cellular glucose uptake, disrupting intracellular balance. Genipin (GP), recognized UCP2 inhibitor, exhibits anti-inflammatory antioxidant properties. study aimed investigate specific role GP periodontal tissue redox signaling mechanism development periodontitis. Methods In this study, we constructed model H O -induced oxidative stress human ligament cells (hPDLCs). vivo , rat periodontitis was established evaluate effects mechanisms alleviating damage tissues cells. Results Cell experiments showed that effectively alleviated dysfunction hPDLCs inhibiting expression function, restoring cell viability, reducing apoptosis. Additionally, intervention increased transporter 4 (GLUT4), thereby uptake. The results animal demonstrated reduced alveolar bone resorption destruction rats periodontitis, inhibited osteoclast differentiation, improved tissue, promoted GLUT4 expression, levels Discussion regulates maintaining homeostasis, enhancing playing central role.

Trigonelline (TRIG) is a natural compound in an alkaloid family found diverse plants. This exerts anti-inflammatory, anti-allergic, anti-oxidative and anti-fibrotic activities several disease models. However, its beneficial role endothelial injury, especially induced by diabetes, unclear. We, therefore, evaluated the effects of TRIG on cellular proteome human (EA.hy926) cells followed functional validation high-glucose (HG)-induced deteriorations. Label-free quantification using nanoLC-ESI-Qq-TOF MS/MS revealed 40 downregulated 29 upregulated proteins TRIG. Functional enrichment analysis DAVID REVIGO tools suggested involvement these altered biological processes molecular functions, particularly cell-cell adhesion, ATP metabolic process, cell redox homeostasis, cadherin binding, hydrolysis activity. Experimental showed that HG triggered endothelial-to-mesenchymal transition (EndMT) (as demonstrated increased spindle index mesenchymal markers, i.e., fibronectin vimentin, decreased PECAM-1 VE-cadherin), oxidized proteins, reduced intracellular ATP, active mitochondria, tube/mesh formation VEGF secretion. successfully abolished all defects HG. These data indicate prevents HG-induced EndMT, oxidative stress, mitochondrial dysfunction, impaired angiogenic activity cells.

Background Proliferative diabetes retinopathy (PDR) seriously affects the vision of patients. Exploring key genes retinal neovascularization is crucial for developing new biomarkers and therapeutic targets. Objective This study aimed to identify associated with in Diabetic Retinopathy (PDR), intending develop would further our understanding progression diabetic improve patient prognosis. Methods The gene data from 36 samples 45 healthy volunteers or patients were selected GEO DataSets (Gene Expression Omnibus), specifically datasets GSE102485 GSE160310. Utilizing SVA algorithm merge limma package R differentially expressed (DEGs), we conducted a bioinformatic analysis retinopathy. Functional insights gained through DAVID database analyses, while STRING database-derived Protein-Protein Interaction (PPI) networks visualized Cytoscape provided context. Key identified LASSO regression SVM ROC curves assessing their diagnostic value. Single set enrichment (GSEA) enhanced perturbed biological processes pathways, advancing knowledge at genomic level. Results A rigorous yielded comprehensive list 1139 which six pivotal genes—KDM5D, AC007040.11, AC015688.3, NLRP2, GYPC, TMSB4Y—were as central study. These consistently demonstrated high accuracy, each exhibiting an area under receiver operating characteristic (ROC) curve (AUC) exceeding 0.75. Gene Ontology (GO) elucidated primary roles intricate processes, including inflammatory immune responses, T-cell activation, cell apoptosis, angiogenesis. Moreover, Kyoto Encyclopedia Genes Genomes (KEGG) pathway revealed involvement signaling cascades such cytokine-cytokine receptor interactions, adhesion molecule PI3K-Akt signaling, hematopoietic lineage, substantiating significance pathogenesis Conclusions In this research, expression without PDR, identifying genes, biomarkers, critical pathways potentially involved PDR progression. Our findings enhance PDR's molecular dynamics offer potential avenues improved prognostication intervention, despite limitations due sample group heterogeneity.
