Abstract Diabetic retinopathy (DR) represents a significant and serious complication associated with diabetes mellitus (DM), often resulting in considerable visual impairment or even blindness. The intricate pathological processes underlying DR complicate the effectiveness of current treatment modalities. Studies have highlighted potential natural products via several beneficial effects including anti-inflammatory, antioxidant, anti-neovascular, anti-apoptotic properties. Flavonoids, saponins, saccharides, alkaloids exhibited various vivo vitro studies. However, clinical utilization these compounds is hindered by issues such as inadequate specificity, low bioavailability, toxicity. Therefore, there pressing need for rigorous studies to confirm efficacy preventing mitigating progression DR.

Introduction Periodontitis is a chronic inflammatory disease closely associated with mitochondrial dysfunction. Uncoupling protein 2 (UCP2), located in the inner membrane of mitochondria, reduces potential (MMP) and adenosine triphosphate (ATP) synthesis by promoting proton leakage across membrane. This leads to decreased energy metabolism efficiency, impairing cellular glucose uptake, disrupting intracellular balance. Genipin (GP), recognized UCP2 inhibitor, exhibits anti-inflammatory antioxidant properties. study aimed investigate specific role GP periodontal tissue redox signaling mechanism development periodontitis. Methods In this study, we constructed model H O -induced oxidative stress human ligament cells (hPDLCs). vivo , rat periodontitis was established evaluate effects mechanisms alleviating damage tissues cells. Trigonelline (TRIG) is a natural compound in an alkaloid family found diverse plants. This exerts anti-inflammatory, anti-allergic, anti-oxidative and anti-fibrotic activities several disease models. However, its beneficial role endothelial injury, especially induced by diabetes, unclear. We, therefore, evaluated the effects of TRIG on cellular proteome human (EA.hy926) cells followed functional validation high-glucose (HG)-induced deteriorations. Label-free quantification using nanoLC-ESI-Qq-TOF MS/MS revealed 40 downregulated 29 upregulated proteins TRIG. Functional enrichment analysis DAVID REVIGO tools suggested involvement these altered biological processes molecular functions, particularly cell-cell adhesion, ATP metabolic process, cell redox homeostasis, cadherin binding, hydrolysis activity. Background Proliferative diabetes retinopathy (PDR) seriously affects the vision of patients. Exploring key genes retinal neovascularization is crucial for developing new biomarkers and therapeutic targets.
