Abstract Objective: To identify potential genetic risk factors for sleep disorders (SD) and to explore the causal associations between lifestyle clinical features with SD, given limitations of traditional observational studies. Methods: Data from published genome-wide association studies (GWAS) were analyzed, encompassing 34 17 as exposures influencing SD. These analyses yielded information on 51 4 outcomes. Sleep deprivation (SD) is commonplace in today's fast-paced society. SD a severe public health problem globally since it may cause cognitive decline and even neurodegenerative disorders like Alzheimer's disease. Melatonin (MT) natural chemical secreted by the pineal gland with neuroprotective effects. The purpose of this study was to investigate protective effect mechanism MT on chronic sleep deprivation-induced impairment. A 3-week modified multi-platform method used create rat model. ABSTRACT Aims This study investigated the roles of lateral basal forebrain glial cell line–derived neurotrophic factor (GDNF) signaling and cholinergic neuron activity, apoptosis, autophagy dysfunction in sleep deprivation–induced increased risk chronic postsurgical pain (CPSP) mice. Methods Sleep deprivation (6 h per day from −1 to 3 days postoperatively) was administered mice receiving skin/muscle incision retraction (SMIR) determine whether perioperative induces mechanical thermal hypersensitivity, increases pain, causes changes neurons activity (c‐Fos immunostaining), apoptosis (cleaved Caspase‐3 expression), (LC3 p62 expression) GDNF expression. Adeno‐associated virus (AAV)‐GDNF microinjected into see expression could reverse duration autophagy. Cholinergic were further depleted by mu p75‐SAP examine pain‐prolonging effects still exist. Results Perioperative enhanced sensation prolonged SMIR mice, which accompanied decreased expression, substantia innominata (SI), magnocellular preoptic nucleus (MCPO), horizontal diagonal band Broca (HDB) (hereafter forebrain). Normalizing AAV‐GDNF inhibited mitigated maintenance. Mice with selective lesion resistant pain‐enhancing prolonging pain‐alleviating therapy. Conclusions promotes chronicity possibly through decreasing causing neuronal dysfunction.
