Rheumatoid arthritis (RA) is a chronic autoimmune disease that primarily manifests with symptoms such as heat and toxin. ABSTRACT Rheumatoid arthritis (RA) is a severe inflammatory autoimmune disease with metabolic changes. RA patients have abnormalities in glycolysis, amino acid metabolism, choline and fatty synthesis. The differential metabolites individuals of animal models were explored to find the potential biomarkers for risk prediction, diagnosis, prognosis perspective metabolism. Moreover, we discussed changes related after treatment anti‐rheumatic drugs, Traditional Chinese Medicine (TCM) explore promising metabolites. In addition, immunological mechanism TCM from metabolism was also clarified. For perspectives research application beneficial clinic, relevant technologies focuses future studies field been proposed accordingly.

Ermiao wan (EMW) is a classical traditional Chinese medicine formula, with two modified versions including Sanmiao (SMW) and Simiao (FMW). These categorized formulas (ECFs) are traditionally used to treat gouty arthritis hyperuricemia. However, their potential benefits mechanisms on diabetes remain be explored. This study aims investigate the overall effects biological differences of ECFs in high fat diet (HFD)-fed mice based network pharmacology transcriptomics. significantly reduced body weight, improved oral glucose tolerance, decreased accumulation, lowered serum insulin inflammatory cytokine levels HFD-fed mice. FMW had better efficacy than EMW SMW. Network analysis revealed that targeted functional modules associated chronic inflammation, lipid metabolism, metabolism. Transcriptome results also showed could inhibit genes inflammation upregulated some Comprehensive QPCR verification indicated beneficial might attributed regulation Ddit3, Ccl2, Esr1, Cyp7a1. provides theoretical basis for clinical use ECFs.
