Hyperuricemia is a metabolic disease attributed to sustained dysregulation of purine metabolism, manifesting as consistently elevated blood uric acid levels. Hyperuricemic nephropathy (HN) renal complication hyperuricemia. It characterized by the deposition urate crystals, inflammatory cell infiltration and tubulointerstitial injury. Ligusticum cycloprolactam (LIGc) novel monomeric derivative active ingredient ligustilide (LIG) from Angelica sinensis (Oliv.). LIG demonstrates anti-inflammatory antioxidant properties. Nevertheless, therapeutic potential LIGc ameliorate HN required further investigation. Our study revealed that effectively reduced serum attenuated in mice induced co-administering potassium oxonate hypoxanthine. research demonstrated treatment improved function with regulating expression transporters. Histopathological analysis showed tubular damage, interstitial collagen deposition. Mechanistically, alleviated injury inhibiting TLR4/NF-κB signaling pathway both vivo vitro. mitigated attenuating response through pathway, providing new perspectives for HN.
