The ApoE4 allele is the strongest genetic determinant for Alzheimer's disease (AD), while obesity a strong environmental risk AD. modulatory effect of ApoE genotype on aging-related cognitive function in tandem with high-fat diet (HFD) remains uncertain. This study aimed to elucidate effects ApoE3/ApoE4 genotypes aged mice exposed HFD, and benefits n-3 polyunsaturated fatty acids (PUFAs) from fish oil. Remarkably, HFD led weight gain lipid accumulation, more pronounced ApoE3 mice, experienced exacerbated cerebral insulin resistance, neuroinflammation, oxidative stress. Critically, PUFAs modulated signaling via IRS-1/AKT/GLUT4 pathway, mitigated microglial hyperactivity, reduced IL-6 MDA levels, thereby counteracting deficits. Background/Objectives: This research aimed to investigate phospholipid fatty acid (PLFA) distribution in the brain, kidneys, and white adipose tissue (WAT) lipid profiles response high-fat diets. Methods: Adult female C57BL/6 mice were fed diets containing 25% linseed, palm, or sunflower oil for 100 days. The composition of dietary oils PL analyzed using gas-liquid chromatography. Results: Linseed increased n-3 polyunsaturated acids (PUFAs) with subsequent conversion into long-chain PUFAs brain kidney PL, while only alpha-linolenic was elevated WAT. Obeticholic acid (OCA) has been examined to treat non-alcoholic steatohepatitis (NASH), but unsatisfactory antifibrotic effect and deficient responsive rate in recent phase III clinical trial. Using a prolonged western diet-feeding murine NASH model, we show that OCA-shaped gut microbiota induces lipid peroxidation impairs its anti-fibrotic effect. Mechanically, Bacteroides enriched by OCA deconjugates tauro-conjugated bile acids generate excessive chenodeoxycholic (CDCA), resulting liver ROS accumulation. We further elucidate reduces triglycerides containing polyunsaturated fatty (PUFA-TGs) levels, whereas elevates free PUFAs phosphatidylethanolamines PUFA (PUFA-PEs), which are susceptible be oxidized peroxides (notably arachidonic (ARA)-derived 12-HHTrE), inducing hepatocyte ferroptosis activating hepatic stellate cells (HSCs).
